Morpholinium toluene-4-sulphonate is a liquid consisting of toluene-4-sulphonic acid (49.5 %), morpholine (25.5 %), and water (25 %).

In an acute inhalation hazard test, a slight irritation of the respiratory tract was detected at vapour-saturated air after 8 hours of exposure. The applied concentration was 5.84 mg/L (= 5840 mg/m3).

In case of taking this study for the derivation of the inhalative DNEL, the starting point had to be modified according to Chapter R.8 of the REACH Guidance. Here, a correction for activity driven differences of respiratory volumes in workers compared to workers in rest should be added to receive a starting point of 3912.8 mg/m3. Additionally, an overall assessment factor of 20 should be established: 5 for intraspecies variability and 2 for dose response and 2 for the validity of data source. No time extrapolation is added since local irritation is a dose-dependent effect, occurs immediately and is therefore not time-dependent. No interspecies factor would be needed due to the already modified starting point. Thus, applying these assessment factors, a health-based occupational exposure limit of 195.64 mg/m3 would be obtained for Morpholinium toluene-4-sulphonate.

But a German occupational exposure limit (OEL) exists for morpholine. This OEL is set at 36 mg/m3 with a maximum exceeding factor of 2. Additionally, the portion of morpholine which comes up to 25.5 % should be taken into account and thus would lead to a DNEL of 144 mg/m3 (factor of 4).

But a value of 144 mg/m3 definitely exceeds the recommended general nuisance level. However, the general nuisance level for dust exposure is 10 mg/m³. This level is considered to be sufficiently protective from systemic effects as well as from local irritation resulting from possible exposure to morpholinium toluene-4-sulphonate.

For that reason, a DNEL of 10 mg/m3 is recommended for morpholinium toluene-4-sulphonate.

There are only industrial uses for morpholinium toluene-4-sulphonate. General population is not exposed via inhalation, dermal or oral route. Therefore no DNEL is calculated for the general population.
