- Test Item Solution
FAT 40825/A (10.27 mg) was dissolved in a solvent mixture of water and methanol (45:55; v/v) (20 ml) to prepare a stock solution of 513.5 µg/ml. 1 ml of this solution was diluted with the same solvent mixture to 10 ml to obtain a standard solution of 51.35 µg/ml. The pH value of the test item solution was determined to be 7.0. This test item solution was injected three times. This method utilizes a high-performance liquid Chromatograph with UV/VIS detector, reversed phase column and an aqueous mobile phase.
Apparatus: Varian autosampler ProStar 410; Varian pump ProStar 230; Varian UVA/IS detector ProStar 310; Jones column oven 7990

- Remarks on result:
- other: HPLC method
- Details on results (HPLC method):
- The chromatography of the test item resulted in one main peak with a well-defined and reproducible retention time. The HPLC method was therefore considered to be suitable for the purpose of the study. Based on the chromatographic data, FAT 40825/A was considered to be stable during the test procedure.
- Validity criteria fulfilled:
- yes
- Conclusions:
- The log of the adsorption coefficient (Koc) of FAT 40825/A was estimated to be log Koc < 1.25 which is equal to a Koc value of < 18 using the HPLC method. This value indicates that FAT 40825/A will not be adsorbed by organic carbon in soil.
- Executive summary:
The determination of the adsorption coefficient (Koc) of FAT 40825/A was performed according to the OECD Guideline for the Testing of Chemicals, No. 121, Estimation of the Adsorption Coefficient (Koc) on Soil and Sewage Sludge using High Performance Liquid Chromatography (HPLC), January, 2001 and the EC Directive 2001/59, C.19. The test item solution was injected three times and the reference solution was injected six times. FAT 40825/A was not retained under the chromatographic conditions of the test and was eluted at the dead time. The Koc of FAT 40825/A was therefore determined to be lower than that one of the lowest reference item used (acetanilide). The chromatography of the test item resulted in one main peak with a well-defined and reproducible retention time. The HPLC method was therefore considered to be suitable for the purpose of the study. Based on the chromatographic data, FAT 40825/A was considered to be stable during the test procedure. In conclusion, the log of the adsorption coefficient (Koc) of FAT 40825/A was estimated to be log Koc <1.25 which is equal to a Koc value of <18 using the HPLC method. This value indicates that FAT 40825/A will not be adsorbed by organic carbon in soil.
