- The saturated stock solution of N,N’- DIPHENYL THIOUREA was obtained by mixing of 100 mg of the test item into 1 L of dilution water, under 3 hours of magnetic stirring, followed by a 0.45 µm filtration (Millipore HAWP04500). The filtrated stock solution was clear and colourless.

- The organisms used for the test were Danio rerio (Teleostei, Cyprinidae). The batch n°11/Br/03/1 was supplied by Elevage de la Grande Rivière France (69490 Saint Forgeux). The fish selected for the test were all from the same batch and were of the same age (date of birth stated by the supplier: 28/06/11). Their size (determined by measuring 10 randomly sampled fish) was 35 ± 4 mm. The mortality in the aquarium containing the batch of fish used for the tests was less than 5 % (observed mortality: 0.7%) over the 7 days preceding the test. The sensitivity of the biological reagent is checked at least once for each new batch of fish, by determining the LC50-24h of the potassium dichromate. For reference, the results of the last test, carried out on 18/10/2011, were a LC50-24h of 220 mg/L (potassium dichromate Merck 1.02403.0080 batch n°102403H). For reference, the former AFNOR standard NF T 90-303 states that the LC50-24h of the potassium dichromate with respect to Danio rerio must be between 200 and 400 mg/L.

- The room was equipped with a programmed lighting system which produced a controlled lighting sequence: intensity between 400 and 800 lux at the surface of the aquariums; photo-period of 12 hours of darkness per 24 h.
The study was performed using aquaria with a nominal capacity of 5 litres, covered with Altuglas® cover plates drilled with holes to allow the test solutions to be aerated. Volume of test medium per aquarium was 2 x 2.5 L.
Physico-chemical parameters were measured using a METTLER TOLEDO 345 pH-meter for measurement of pH and with a WTW OXI 538 oxymeter for dissolved oxygen measurement.
Preliminary test: series of 5 concentrations: 100, 50, 10, 5, 1 % of saturated stock solution of N,N’- DIPHENYL THIOUREA in dilution water.
Five fish were added per aquarium, only one vessel was prepared for each concentration.
The pH and the dissolved O2 were measured at the beginning and at the end of the test, at the highest concentration and in the control vessel.
Definitive test, nominal concentrations tested: 100, 56.2, 31.6, 17.8 and 10 % of saturated stock solution of N,N’- DIPHENYL THIOUREA (factor 1.78).
At the beginning of the test, appropriate volumes of the stock solution of N,N’- DIPHENYL THIOUREA were poured directly into each test aquarium, which was then topped up with dilution water to obtain the above mentioned range of nominal concentrations (no pH-adjustment done).
Number of fish: 10 for each test concentration and for the control.
The definitive test was carried out under static conditions.
