Short term toxicity to fish

One OECD 203 study is available with one the main constituents (Bis (2-chloroethoxy) methane) which is the most toxic. ECOSAR predicted values are available for the three main constituents (including Bis (2-chloroethoxy) methane) and they are as follows:

Bis (2-chloroethoxy) methane 96 hr LC50: 125.22 mg/L (compare to measured value of 50.2 mg/L)

Ethane, 1-(2-chloroethoxy) -2-[(2-chloroethoxy) methoxy: 225.23 mg/L

1,15-dichloro-3,5,8,11,13-penta-oxa pentadecane: 96 hr LC50 = 610.849 mg/L

The predicted data show that Bis (2-chloroethoxy) methane is the most toxic of the three main components.

As a result the measured value of 50.2 mg/L is taken as the worst case and used for risk assessment purposes, without a need to perform full tests with the remaining 2 components or the complete substance mixture.

Long term toxicity to fish

Test waived on the basis of the following. 1. strictly controlled conditions - exposure assessment is waived. 2. Acute to chronic ratio - looking at the data for Daphnia and algae, the acute to chronic ratios are Daphnia (31.7 mg/L/20 mg/L = 1.6) and algae (253.6 mg/L/81.3 mg/L = 3.1). These figures are very low (< 10) indicating that there is not that much difference between acute and chronic toxicity, showing that acute toxicity is the main mechanism of toxicity and is sufficient to represent the toxicity of the substance. 3. For ethical/animal welfare reasons, conducting an animal test would not add any additional value to the risk assessment process due to reason 2 above. 4. ECOSAR predicts 96 hr EC50 to fish for Bis (2-chloroethoxy) methane (125 mg/L) comparable to the measured value of 50.2 mg/L Predicted toxicity to fish of the other 2 main components  (96 hr EC50: 610 mg/L and 225 mg/L) are all higher than that of Bis (2-chloroethoxy) methane. This supports the fact that acute toxicity to Bis (2-chloroethoxy) methane was used as a worst case in the dossier.

Short term toxicity to aquatic invertebrates

One short-term study (OECD 202), valid without restrictions, with Daphnia magna is available. The test was performed under GLP with a static setup. All measured test concentrations were close to nominal, therefore effects were based on nominal test concentrations. The 48h-EC50 is 31.7 mg/L (nominal, test material).

Long term toxicity to aquatic invertebrates

Based on a 13-day range finding test in M4 medium with no effect up to the highest test concentration of 10 mg/L (nominal) the following nominal test concentrations were prepared: 1.0, 2.2, 4.5, 9.5 and 20.0 mg/L for a full OECD 211 test. All measured test concentrations were close to nominal, therefore effects were based on nominal test concentrations.

The 21 d NOEC (reproduction, parent length and weight) was > 20 mg/L

Toxicity to aquatic algae and cyanobacteria

One study (OECD 201), valid without restrictions, with freshwater algae P. subcapitata is available. The test was performed under GLP. All measured test concentrations were close to nominal, therefore effects were based on nominal test concentrations.

The 72h-ErC10 is 81.8 mg/L (nominal, test material).

The 72h-ErC50 is 254 mg/L (nominal, test material).

Toxicity to microorganisms

One OECD 209 test is available. The EC50 of Reaction mass of 1,15-dichloro-3,5,8,11,13 -pentaoxapentadecane and 1-chloro-2-[(2-chloroethoxy)methoxy]ethane and 1 -chloro-2-{[2-(2-chloroethoxy)ethoxy]methoxy}ethane for activated sludge after 3 hours contact time is >1280 mg/L.
