Description of key information
Skin irritation test performed in vivo :
The relative level of primary skin irritation of the test sample "zinc gluconate granular" was investigated in one group three rabbits. The exposition period of the test substance was 4 hours. Any signs of erythema, eschar and edema formation were recorded during the observation period of 72 hours. The judgement of the skin alterations was done 30' - 60', 24h, 48h and 72h p.a. after the patch removal. The sample did not induce any skin irritation during the entire observation period of 72 h. The primary irritation index was 0.0. All animals showed a normal food consumption and a normal weight gain. The sample "zinc gluconate granular" is considered to be not irritating under the conditions described.
Eye irritation test performed in vivo:
An in vivo eye irritation test was conduct on 6 rabbits (New Zealand white). The eyes were scored at 1, 2, 8 hours and 1, 2, 3, 4, 5, 6 & 7 days after the treatment. The untreated eye serves as the control, as recommended by the OECD Guideline. No cornea opacity was observed. The maximum iris score was 1. The maximal conjunctivae score was 3. The maximal chemosis score was 3. All the effects observed were fully reversible (within four days maximum). The substance is considered slightly irritant in this study.
A second in vivo study was performed on 3 rabbits (New Zeland white). The substance tested (zinc gluconate) is considered irritant to eyes with risk of serious damage. Considering the irreversible effects observed during the study period, the substance shloud be considered in category 1 based on GHS criteria.

- Irritant / corrosive response data:
- One hour after instillation of the substance, reactions are noticed in all animals at the level of the conjunctivae (diffuse purple enanthemae, lacrimations and chemosis) and at the level of the iris (congestion). 24 hours later, some of these reactions increase ; a comeal opacity is also noticed in the three rabbits. 72 hours after instillation, the mean conjunctival irritation signs have disappeared for one rabbit, excepted enanthema that is no more visible at the day 12.
- Interpretation of results:
- Category 1 (irreversible effects on the eye) based on GHS criteria
- Conclusions:
- The substance tested (zinc gluconate) is considered irritant to eyes with risk of serious damage. Considering the irreversible effects observed during the study period, the substance shloud be considered in category 1 based on GHS criteria.
- Executive summary:
One hour after instillation of the substance, reactions are noticed in all animals at the level of the conjunctivae (diffuse purple enanthemae, lacrimations and chemosis) and at the level of the iris (congestion). 24 hours later, some of these reactions increase ; a comeal opacity is also noticed in the three rabbits. 72 hours after instillation, the mean conjunctival irritation signs have disappeared for one rabbit, excepted enanthema that is no more visible at the day 12.
For the two other animals, all irritation signs, excepted iris congestion, are still visible at the end of the observation period.

Endpoint conclusion
- Endpoint conclusion:
- adverse effect observed (irreversible damage)
Respiratory irritation
Endpoint conclusion
- Endpoint conclusion:
- no study available
Additional information
Justification for classification or non-classification
No skin irritation is observed in an in vivo study.
Two in vivo studies are available. The most recent study was considered considering that it is more reliable and that it correspond to a conservative approach. In this study, the substance is irritant and cause irreversible damage. This correspond to the classification as eye irritant category 1 (H318: cause serious eye damage).
