Description of key information

Adequate reliable measured data exists for short-term toxicity to fish DMDMH and has been documented within this dossier. The most sensitive reliable study result has been identified as a study (Ciliutti 2003) which reports an LC50 for short term toxicity in fish of >82.3 mg/L. Consequently this value will be taken as the short-term toxicity in fish endpoint for DMDMH.

Additional information

Reliable (Klimisch 1 or 2) short-term toxicity studies with fish have been conducted for DMDMH or the hydrolysis product of DMDMH, DMH and are included in this dossier. Studies conducted with the hydrolysis product, DMH, are considered relevant for environmental fate and ecotoxicology and are considered in this dossier.

 

Ciliutti (2003) conducted a reliable (Klimisch 1) GLP compliant study following methods similar to OECD 203. Zebrafish (Brachydanio rerio) 2-3cm long, were exposed to measured concentrations (82.3, 33.3, 14.2, 7.2, 2.6 mg/l) of DMDMH in a semi-static system for 96 hours. The 96-hr LC50, based on mortality, was >82.3 mg/L. 

 

Murphy (1992) conducted a reliable (Klimisch 1) GLP compliant study following methods similar to OECD 203. Rainbow trout (Oncorrhynchus mykiss) juveniles were exposed to one measured concentration (972 mg/L) of the hydrolysis product of DMDMH, DMH, in a static limit test. The 96-hr LC50, based on mortality, was >972.2 mg/L. 

 

Department of Clinical and Industrial Toxicology of Industrial Biotest Laboratories (1982) conducted a reliable (Klimisch 2) study without following a specified guideline. Bluegill sunfish (Lepomis macrochirus) 3.5-7.5 cm long, were exposed to nominal concentrations (100, 135, 180, 240 and 320 mg/l) of DMDMH in a static system for 96 hours. The 96-hr LC50, based on mortality, was 173 mg/L. 

 

Department of Clinical and Industrial Toxicology of Industrial Biotest Laboratories (1982) conducted a reliable (Klimisch 2) study without following a specified guideline. Rainbow trout (Oncorrhynchus mykiss) 3.5-7.5 cm long, were exposed to nominal concentrations (240, 320, 420, 560, and 750 mg/l) of DMDMH in a static system for 96 hours. The 96-hr LC50, based on mortality, was 283 mg/L.
