In Article 13 of Regulation (EC) No 1907/2006, it is laid down that information on intrinsic properties of substances may be generated by means other than tests, provided that the conditions set out in Annex XI (of the same Regulation) are met.

 

According to Annex XI of Regulation (EC) No 1907/2006 (Q)SAR results can be used if (1) the scientific validity of the (Q)SAR model has been established, (2) the substance falls within the applicability domain of the (Q)SAR model, (3) the results are adequate for the purpose of classification and labeling and/or risk assessment and (4) adequate and reliable documentation of the applied method is provided.

 

For the assessment of 3,4 -dimethoxyphenethylamine (CAS 120 -20 -7) (Q)SAR results were used for toxicity to aquatic organisms. The criteria listed in Annex XI of Regulation (EC) No 1907/2006 are considered to be adequately fulfilled and therefore the endpoint(s) sufficiently covered and suitable for risk assessment.

 

Therefore, experimental studies on aquatic toxicity are not provided.

The assessment of the short-term toxicity of 3,4 -dimethoxyphenethylamine (CAS 120-20-7) to aquatic invertebrates is based on a read-across approach to the two structurally analogous substances and is supported by estimates from several QSAR model for the target substance itself.

Both compounds were tested on Daphnia magna according OECD 201. A study with 3,4 -dimethoxyphenylacetonitrile (CAS 93-17-4) resulted in a 48-h EC50 of greater than 100 mg/L. All nominal test concentrations were analytically verified (BASF AG, 2003, report no. 02/0651/50/1).

4 -allylveratrole appears to be more toxic with a 48-h EC50 of 38 mg/L (meas.). The data are from a reliable source (NITE, J-CHECK database, Japan). Though no test report is available at present the data from this source are known to be well documented and the test are performed according to GLP and OECD guidelines.

In addition, the short-term toxicity was calculated for the test substance 3,4 -dimethoxyphenethylamine (CAS 120-20-7) with several estimation models. The table below lists the result from the described above OECD studies with the structurally analogous substances 4 -allylveratrole (CAS 93-15-2) and 3,4 -dimethoxyphenylacetonitrile (CAS 93-17-4) and, in addition, the applied (Q)SAR models, the estimated effect toxicity and information on the applicability domain (AD) for the target substance.

The target substance 3,4 -dimethoxyphenethylamine (CAS 120-20-7) is estimated to be acutely harmful to aquatic invertebrates based on the models where the substance was within the applicability domain, except for the model ECOSAR v1.11 (48 -h LC50 = 338 mg/L). According to the VEGA models, the substance would be acutely toxic to aquatic invertebrates. However, the substance was not within the applicability domain.

Following a worst-case assessment, 3,4-Dimethoxyphenethylamine (CAS 120-20-7) is considered to be acutely harmful to aquatic invertebrates.
