According to the data provided in the REACH dossier of butanone, dissiminated on the ECHA homepage, butanone is not mutagenic in bacteria, mammalian cells in vitro or in mammals.

AMES (S. typ. TA 1535, TA 1537, TA 1538, TA 98, TA 100): negative

Mouse lymphoma assay (L5178Y cells): negative

Chromosomal abberation test in vitro (rat liver RL4 cell line): negative

Unscheduled DNA synthesis test in vitro(rat hepatocytes): negative

Cell transformation assay (BALB/3T3): negative

Micronucleus test in vivo (mouse): negative

Further data from secondary sources (MAK documentation of the Deutsche Forschungsgemeinschaft-DFG) reported, that butanone has been tested negative in E. coli strains WP2, WP2uvrA and in S. typ. strain TA102.
