Study design

Oxygen conditions:
anaerobic
Inoculum or test system:
natural sediment
Details on source and properties of sediment:
The sediment used in the experiment was collected at the mouth of Tsurumi River Japan. The surface sediment sample was collected by an Ekman-Barge sediment sampler from the Tsurumi-Ohashi bridge in September 1992.
The sediment was black in color indicating sulfate reducing activity.
Macro benthic organisms, such as crabs, small fish and shellfish were present in the collected sediment.
The sediment was sieved through a 2 mm screen to remove debris.
The sieved sediment sample was mixed with river water to make sediment slurry with a solids concentration of 272 +/- 2.8 g/Kg.
This was an average of four replicate measurements.
The pH of the sediment slurry was 5.6.
Duration of test (contact time):
1 yr
Initial test substance concentration
Initial conc.:
4 µmol/L
Based on:
test mat.
Details on study design:
- for each replicate a 5mL of sediment slurry was placed in a screw top test tube and sealed under N2 atmosphere.
- the tubes were sealt and kept at room temperature for one week to ensure that the system was anaerobic.
- all test tubes were placed in an anaerobic chamber (10% H2, 10% CO2, 80% N2)
- 10 µL of stock solution was added to each test tube to give an initial concentration of 4 µmoles/L
- immediately after spiking, time zero (Co) samples were frozen at -20 °C, while other test tubes were kept in a constant temperature room at 25 °C
- the slurries were hand mixed three times a week by turning upside down for few minutes
- the test tubes were sampled once a day at the beginning of the incubation then at longer time intervals as the experiment progressed
- the incubation lasted for a year
