Description of key information

Three published data sources report that 2-chloropropionic acid is corrosive to skin. One source reports that the sodium salt of 2-chloropropionic acid is non irritating in a study conducted according to OECD guideline 404 under GLP.
Two published studies in the IUCLID 4 dataset report that 2-chloropropionic acid is irritating to eyes and one study reports that the sodium salt of 2-chloropropionic acid is irritating to eyes. An internet source states that human evidence indicates the substance to be corrosive to eyes.

Additional information

Three published data sources, one from American Conference of Governmental Industrial Hygienists and two from the IUCLID 4 dataset report 2-Chloropropionic acid to be corrosive. These three sources together constitute an assessment of the available human and animal data - the American Conference of Governmental Industrial Hygienists indicates that moderate skin burns have been observed in workers with acute exposure to the liquid. No irritation was noted in employees exposed to the vapour at TWA concentration up to 0.35 ppm with occasional exposure up to 1.95 ppm.

A fourth IUCLID 4 dataset entry reports that the sodium salt of 2-chloropropionic acid was non irritating when tested according to OECD guideline 404. It is suggested that there was little dissociation of the salt in this study and thus insignificant quantities of the free acid were available during the study in order to result in a corrosive outcome.

As regards eye irritancy, two IUCLID 4 dataset endpoints report that 2 -chloropropionic acid is irritating to eyes and a study conducted according to guideline OECD 405 (to GLP) reports that the sodium salt of 2-chloropropionic acid was also irritating to eyes. In addition, an internet source states that human evidence indicates the substance to be corrosive to eyes.

Justification for classification or non-classification

The studies summarised above suggest that the substance should be classified as corrosive to skin and irritating to eyes.

In addition, the substance is listed in Annex I of Directive 67/548/EEC as Corrosive, C, (R35 : Causes severe burns) and in Annex VI (Table 3.1) of CLP-Regulation (EC) No 1272/2008 as Skin Corr. 1A (H314: Causes severe skin burns and eye damage).
