Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Results and discussion

Results of examinations

Clinical signs:
effects observed, treatment-related
Description (incidence and severity):
All female rats in the 5000 mg/kg group subsequently died. Signs of neurotoxicity were seen.
Mortality:
mortality observed, treatment-related
Description (incidence):
All female rats in the 5000 mg/kg group subsequently died. Signs of neurotoxicity were seen.
Details on results:
Clinical signs indicative of neurotoxic effects (tremors, hyperreflexia, twitching of extremities and convulsions (i.e., opisthotonos)) were observed in female rats after a single oral dose of DMBC in corn oil at 5000 mg/kg (10 mL/kg). All female rats in the 5000 mg/kg group subsequently died. One male rat in the 5000 mg/kg group showed tremors; however, this animal survived. No other males showed neurotoxic signs.

Applicant's summary and conclusion

Conclusions:
DMBC showed neurotoxic effects in an acute oral study conducted in rats.
Executive summary:

Clinical signs indicative of neurotoxic effects (tremors, hyperreflexia, twitching of extremities and convulsions (i.e., opisthotonos)) were observed in female rats after a single oral dose of DMBC in corn oil at 5000 mg/kg (10 mL/kg). All female rats in the 5000 mg/kg group subsequently died. One male rat in the 5000 mg/kg group showed tremors; however, this animal survived. No other males showed neurotoxic signs.

The oral LD50 was between 2000 and 5000 mg/kg in females and greater than 5000 mg/kg in males. No clinical signs indicative of neurotoxicity were observed at doses of 2000 mg/kg or less in either sex.
