EC50 nominal (48 h) = 24.8 mg/L for Daphnia magna (EU Method C.2 of directive 92/69/EEC)

In a GLP study according to Directive 92/69/EEC, C.2, with analytical monitoring, the EC50 of Daphnia magna was 24.8 mg/L (nominal) after 48 h (Bayer AG, 2003). Due to rapid hydrolysis of dimethyl phosphonate to monomethyl phosphonate, and the slow hydrolysis of monomethyl phosphonate to phosphonic acid, there were significant concentrations of monomethyl phosphonate at any dimethyl phosphonate concentration (22 - 35% w/w of initial dimethyl phosphonate). At the nominal dimethyl phosphonate concentration of 25 mg/L, the geometric mean of the measured monomethyl phosphonate concentrations was 7.9 mg/L after 48 h whereas dimethyl phosphonate was not detectable anymore at this time. DOC measurements confirmed the presence of virtually all organic carbon of the initial carbon content of dimethyl phosphonate.
