Description of key information

Condensation product of N-C12-C18-alkylpropane-1,3-diamine, N-(3 -aminopropyl)-N’-C12-C18-alkylpropane-1,3-diamine and formic acid exhibit acute toxicity to aquatic micro-organisms.

The effect of “Condensation product of N-C12-C18-alkylpropane-1,3-diamine, N-(3 -aminopropyl)-N’-C12-C18-alkylpropane-1,3-diamine and formic acid"

on the respiration rate of activated sludge was assessed by the methods detailed in EC Regulation 440/2008, Method C.11, corresponding to OECD Test Guideline 209, ‘Activated Sludge, Respiration Inhibition Test’ (1981). The study (Dickinson 2010) is considered valid and conclusive.

Samples of activated sludge (suspended solids 1.6 g/L), fed with synthetic sewage, were exposed to dilutions of the test substance for three hours. Their rates of oxygen consumption were determined using an oxygen electrode and were compared with those of controls that contained activated sludge and synthetic sewage alone and were established at the beginning and end of the test.

The specific respiration rate of the control culture established at the end of the test (Control 2, 34.9 mgO2/g/h) was 98% of the rate established at the start (Control 1, 35.7 mgO2/g/h). The three-hour 50% effect concentration (EC50) for 3,5-DCP was calculated to be 9.9 mg/L (95% confidence limits, 8.5 to 11.6 mg/L). These results show that the sample of activated sludge employed was sensitive to inhibition.

In the sample of activated sludge at a nominal test item concentration of 10 mg/L, no significant inhibition of the respiration rate was observed (NOEC). Concentration-related inhibition of respiration rates was observed at nominal concentrations between 30 and 300 mg/L; rates were decreased by 15% to 98%. At 1000 mg/L (triplicate) the respiration rates were decreased by 98, 99 and 97%, and were similar to that measured at 300 mg/L.

The three-hour EC50 was calculated to be 75.2 mg/L (95% confidence limits, 66.8 to 84.3 mg/L). The EC20 and EC80 were calculated to be 37.4 and 151 mg/L (95% confidence limits, 30.3 to 46.4 mg/L and 132 to 175 mg/L), respectively.
