Pyridine, the sentinal member of the pyridine and methylpyridine category, is noted as not sensitising in a reliable toxicology text, Patty's Industrial Hygiene and Toxicology, New York, NY: John Wiley & Sons Inc., 1991. Pyridine was included in the validation data set for the ICCVAM Local Lymph Node Assay, and was found to be a weak sensitiser. The authors of the ICCVAM study noted that this positive result was unexpected, since it was considered not to be a sensitising substance.

A category of pyridine and methyl pyridine derivatives is comprised of: pyridine, 2-methylpyridine, 3-methylpyridine and 4-methylpyridine. The basis of the category is structural similarity (based on the pyridine unsaturated ring structure) and similar physical properties, environmental fate and ecotoxicity, and mammalian toxicity. Similar toxicological properties derive from similar physical-chemical properties and commone pathways of metabolism and elimination among all members of the category. This category is accepted by the U.S. Environmental Protection Agency (EPA).

Justification for classification or non-classification

2 -Methylpyridine is listed in Regulation (EC) No. 1272/2008, Annex VI, Index # 613-036-00-2), and is not classified as a sensitiser. Two other members of the category are also listed as non-sensitisers: Pyridine (Index #613-002-00-7, and 4-methylpyridine (Index # 613-037-00-8). Based on the absense of firm data to the contrary, the Annex VI classification is retained, and 2 -methylpyridine is not classified as a sensitiser.
