- Executive summary:
The density of the substance was determined under GLP in accordance with OECD TG 109, using a gas comparison pycnometer method. This method allows determination of the true volume of solid samples and therefore, through the known mass of sample taken, also the density of the sample. The instrument, which can be pressurized with helium, consists of two interlinked cells (reference cell and sample cell) of fixed volume. Helium gas is used as it is chemically inert and also demonstrates ideal gas behaviour. The volume of the reference cell and sample cell are confirmed prior to test sample analysis through the use of a stainless steel calibration sphere of certified volume. Sample analysis is then performed by recording the pressure drop for the pressurized reference cell when the additional volume of the sample cell containing the test substance is added. As the volume of the reference cell and sample cell (empty) are confirmed during calibration, the volume of test substance present can then be accurately calculated. The instrument was calibrated using reference cells with defined volumes. Aliquots of test item (38.3316 g and 40.8034 g for determination 1 and determination 2 respectively) were weighed into the sample cell and placed into the pycnometer. Triplicate pressure readings (P1(sample) and P2(sample)) were taken after pressurizing the reference cell of known volume (Vref) and then switching to the sample cell (Vcell). The density was then calculated for each pressure reading, and the mean density of 1.65 g/cm3 at 20 °C was calculated from the determinations.
