Description of key information

The primary objective of the OECD 211 is to assess the effect of the total number of living offspring produced per surviving parental animal at the end of test. Therefore the NOEC for this endpoint has been selected as the key value for PNEC derivation and classification.

The effects of 2-methylundecanal on the survival, reproduction and growth of Daphnia Magna was determined during a 21 day exposure period under closed-bottle static-renewal test conditions with a daily renewal of test solutions. Daphnids were exposed to a geometric series of six test concentrations, a negative control (dilution water) and a solvent control (80 µL/L HPLC-grade dimethylformamide). The nominal test concentrations were 18, 30, 50, 85, 150 and 250 µg a.i./L. Time-weighted mean measured test concentrations were determined from samples of test solution collected from each treatment group at the beginning and end of the first and last renewals of the test, and at the beginning and the end of at least one renewal cycle each week during the test. The mean measured test concentrations were 5.8, 7.8, 11, 17, 33 and 59 µg a.i./L, representing 32, 26, 23, 20, 22 and 23% of the nominal test concentrations, respectively. The results of the study are based on the mean measured test concentrations.

Negative control and solvent control data for each parameter were compared using an appropriate statistical test. Since no significant differences between the control groups were found for any parameter tested (p > 0.05), the control data were pooled for comparison with the treatment groups.

There were no statistically significant treatment-related effects on survival, reproduction or growth at concentrations ≤33 µg a.i./L. Survival was the most sensitive biological endpoint measured in this study. Daphnids exposed to 2-methylhendecanal at a concentration of 59 µg a.i./L had statistically significant reduction in survival in comparison to the pooled control. Consequently, the overall NOEC and LOEC for the study were determined to be 33 and 59 µg a.i./L, respectively.
