As the test substance is amphoteric (amino and carboxylic groups), it is not possible to generate nonionic form according to EG guideline 92/69 A.8. The pH 12 was chosen, because at this pH both amino functions are nonionic, the carboxylic acid function is definitely deprotonated, but probably compensated by sodium ions
