Description of key information
Two acute fish toxicity tests were conducted with 1-nitropropane according to the OECD 203 fish toxicity guideline. The most recent test (2009) was conducted under GLP using fathead minnows (Pimephales primelas) while the older test was a non-GLP study using Zebra fish (Brachydanio rerio (new name: Danio rerio)). In addition, a QSAR value for fish was generated using the USEPA ECOSAR v1.00 model as part of EPISuite v4.
The key study for fish toxicity is the most recent fathead minnow test. This test was conducted under GLP, using flow-through conditions to maintain test concentrations due to the volatile potential for the chemical. Analytical verification of test solutions was conducted and demonstrated that test concentrations were on target and remained stable for the duration of the exposure. The results are calculated based on mean measured concentrations. The 96-hour LC50 was 227 mg 1-nitropropane/L.
The supporting study determined a similar value with a 48-hour LC50 of 205 mg 1-nitropropane in the zebra fish (Brachydanio rerio (new name: Danio rerio)).

The report for the original study with the zebra fish did not provide adequate detail on test conditions or test methods to be considered a key study. This study also only reported a 48-hour EC50 although it indicated a 96-hour exposure was conducted. The report indicated that measures were taken to maintain test concentrations via closed-system due to the potential for volatility of the test material but failed to report analytical verification results for test concentration analysis. The reported results from this test were consistent with the results for the recent test (48-hour LC50 values of 205 and 233 mg/L, respectively).
The ECOSAR LC50 value was 582 mg/L, within a factor of three of the experimental values.
These results indicate that 1-nitropropane is not dangerous to freshwater fish species.
