Dilution water for the test was obtained from a well, treated with a Continental Model 3020 Reverse Osmosis Water System, and deionized; then, following the procedures of Marking and Dawson (1973) it was reconstituted to soft water with total hardness of 44 mg/L as CaCO3, total alkalinity of 30 mg/L as CaCO3 and conductivity of 150 micromhos per centimeter. A fresh working stock solution of the test material in deionized water was prepared by weight to a precision of 1.0 mg/L.

Five concentrations were tested, with a control. Testing was begun by adding measured volumes of stock solution to the water in five test vessels, and mixing thoroughly, to yield analyzed test material concentrations of 11.8, 17.1, 28.6, 50.8 and 87.9 mg/L.

The definitive test was conducted in 19.6 liter, chemically clean glass jars, each containing 15 liters of soft reconstituted dilution water and immersed in a constant temperature water bath. Two days before starting the test the fish (Oncorhynchus mykiss) were selected at random from the stock culture, taken off feed, and isolated in a jar of dilution water for 48 hours acclimation before testing. Two test organisms were introduced into each container, mortalities were recorded every 24 hours for 96 hours.

The 96 hour LC50 with 95 percent confidence limits of cyromazine to the rainbow trout (Oncorhynchus mykiss) could not be determined due to the lack of mortalities in all concentrations. The no effect concentration during the test period was observed to be 50.8 mg/L. Behavioral observations (general behavior: norreal, irritated, sounding, surfacing and respiration behaviour: mucus shedding, irregular) were made in the highest concentration of 87.9 mg/L.

Executive summary:

This study was conducted to determine the static acute toxicity of cyromazine to the rainbow trout (Oncorhynchus mykiss). Test practices followed were those recommended by the Committee on Hethods for Toxicity Tests with Aquatic Organisms (1975), except that replicate concentrations were not used.


 


Testing was performed by directly adding the test material to the water in five test vessels, and mixing thoroughly, to yield analyzed test material concentrations of 11.8, 17.1, 28.6, 50.8 and 87.9 mg/L. Ten Oncorhynchus mykiss were introduced at random into each of the test and control vessels. These were approximately 2 months old with a mean length of 40 mm and mean weight of 0.33 grams.


 


The 96 hour LC50 of cyromazine to the rainbow trout (Oncorhynchus mykiss) could not be determined due to the lack of mortalities in all concentrations. The no effect concentration (NOEC) during the test period was observed to be 50.8 mg/L.
