No studies investigating the short-term toxicity to aquatic invertebrates are available for N-(1-oxotetradecyl)sarcosine (CAS 52558-73-3). Therefore, in accordance with Regulation (EC) No. 1907/2006 Annex XI, 1.5 a read-across to the structurally related source substance Sodium N-methyl-N-(1-oxotetradecyl)aminoacetate (CAS 30364-51-3), the respective sodium salt, was conducted.

This read-across approach is in compliance with Annex XI, 1.5, of Regulation (EC) No 1907/2006. The chemical properties of the target and source substance are similar due to the structural similarities, i.e., sarcosine head group and the hydrocarbon tail. In addition, at physiological pH values (above 6) the target substance is expected to be in the anionic form, as are the source substance. As a result, the chemistry of the target and source substance, in solution (above pH 6), are expected to be similar. Thus, the source substance is considered suitable representative for the assessment of the aquatic toxicity of the target substance. A detailed analogue approach justification is provided in the technical dossier (see IUCLID Section 13).

The study conducted with the source substance Sodium N-methyl-N-(1-oxotetradecyl)aminoacetate (CAS 30364-51-3) was performed according to OECD Guideline 202 and GLP. Daphnia magna was exposed to nominal, filtered test concentrations of 1, 3.2, 10, 32 and 100 % v/v saturated solution for 48 h. Analytical measurements of test concentrations using HPLC-MS resulted in significantly lower measured concentrations (1.7 mg/L and 5.1 mg/L) with respect to nominal concentrations (32 and 100 % v/v saturated solution). Thus, the effect concentrations were based on the arithmetic mean measured concentrations. Effects on daphnids mobility were only observed at the highest concentration (5.1 mg/L, measured), leading to an EC50 (48 h) of 2.9 mg/L (arith. mean measured).

Based on the available results from a structurally related read-across substance (in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5), which is characterized by a similar chemistry, it can be concluded that the target substance N-(1-oxotetradecyl)sarcosine (CAS 52558-73-3) will exhibit comparable effects on aquatic invertebrates.
