- The concentration and stability of the test material in the centrifuged and uncentrifuged samples were verified by chemical analysis at 0 (fresh media), 24, 48, 72 (old and fresh media) and 96 hours (old media).

- An amount of test material (100 mg) was dissolved in tetrahydrofuran and the volume adjusted to 25 mL to give a 100 mg/25 mL solvent stock solution. Serial dilutions were prepared from this to give a 0.40 mg/10 mL solvent stock solution. An aliquot (2.0 mL) of the 0.40 mg/25 mL solvent stock solution was dispersed in dechlorinated tap water and stirred with a flat bladed mixer for approximately 1 minute to give the required test concentration. This method of preparation was conducted in duplicate to give replicates R1 and R2.
Each of the stock solutions were inverted several times to ensure adequate mixing and homogeneity.

Current regulatory advice is that in cases where a decline in measured concentrations is observed, time-weighed mean measured concentrations should be used for calculating LC50 values. It was therefore considered justifiable to base the results on the time-weighted mean measured test concentrations in order to give a "worst case" analysis of the data. 0.0034 mg/L was the highest attainable test concentration that could be prepared due to the limited solubility of the test material.

- Based on the time-weighted mean measured test concentrations of the centrifuged test media the acute toxicity of the test material to rainbow trout gave a 96-hour LC50 value of greater than 0.0034 mg/L. The No Observed Effect Concentration was 0.0034 mg/L.
