Short-term toxicity to fish

Description of key information

The acute toxicity of the test material to rainbow trout (Oncorhynchus mykiss) was performed according to OECD TG 203 and in compliance with GLP. The concentration employed in the study was 0.20 mg/L, as this level was established to be the maximum solubility in water. Dimethylformamide was used to spike the test medium. Chemical analysis of the freshly prepared test media at 0, 24, 48 and 72 hours showed measured concentrations to range from 21% to 77% of nominal for the uncentrifuged samples and from 14 % to 50% of nominal for the centrifuged samples. Therefore, the results were based on the time-weighted mean measured test concentrations of the centrifuged test media to give a "worst case" analysis of the data (representing the bioavailable concentration). No mortality or any other effects on the exposed fish were observed. The 96-h LC50 based on the nominal and on the time-weighted mean measured test concentrations of the centrifuged test media was > 0.20 and > 0.055 mg/L, respectively.

Two studies are available. The key study is described at the short key information. The supporting study from Wu (2005) was performed with zebrafish (Brachydanio rerio) using a method comparable to OECD TG 203 and in compliance with GLP. A range-finding test was conducted under semi-static conditions, with concentrations of 10.0 mg/L, 100.0 mg/L and a control group. No mortality or sub-lethal effects were observed at any tested concentrations, but insoluble material was observed in both the 10.0 and 100.0 mg/L test solutions. A definitive test was conducted with a control and a limit test of 100.0 mg/L (nominal concentration). No mortality or sub-lethal effects were observed at the limit tested concentration. Insoluble material was observed in the nominal concentration of 100.0 mg/L test solution. Because the solubility of the test material in water is much lower than 100.0 mg/L, a final definitive test was conducted with the concentration level of 0.20 mg/L. A solvent control group (0.1 mL/L dimethylformamide, DMF) was performed in parallel. No insoluble material was observed in the concentration of 0.20 mg/L test solution. According to the results of the tests above, the 96-hour LC50 was determined to be > 0.20 mg/L.
