Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Endpoint:
short-term toxicity to fish
Type of information:
experimental study
Adequacy of study:
key study
Reliability:
1 (reliable without restriction)
Rationale for reliability incl. deficiencies:
other: GLP guideline study. Since the hydrolysis of the substance is slow, the daily renewal of the test solution is considered sufficient to keep the test concentration within an acceptable range. No effects were observed.

Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION
Initially, 10 mL of test substance was diluted 1:1 with methanol. This mixture was prepared under nitrogen in a glovebox and stored in a freezer prior to use. Weighed amounts with an average of 400.0 ± 1.2 mg from a DEMB/methanol mixture were dissolved in 2000 mL test medium to provide a DEMB concentration of 100 mg/L. The final concentration of methanol in all test solutions was 100 mg/L.

- Biomass loading rate: 0.74 g fish/litre, i.e. 7 fish per 2 litres of test medium.

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: Milli-Ro water (tap.water purified by reverse osmosis; Millipore Corp., Bedford, Mass., USA)
- Culture medium different from test medium: no
- Intervals of water quality measurement: temperature, pH and dissolved oxygen measured daily

OTHER TEST CONDITIONS
- Photoperiod: 16 hours photoperiod daily

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) :
- At 3, 24, 48, 72 and 96 hours following the start of exposure.

TEST CONCENTRATIONS
- Range finding study
- Test concentrations: 0.1, 1, 10, 100 mg/L
- Results used to determine the conditions for the definitive study: No mortality occured during the 96-hour exposure period. However, fish exposed to concentrations of 1 to 100 mg/L showed slight hypoactivity from 24 hours of exposure till the end of the test.
Reference substance (positive control):
yes
Remarks:
pentachlorophenol

Effect conc.:

Nominal / measured:

Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
- No mortality occurred during the 96-hour exposure period. Further, fish did not show any effect on swimming behaviour or appearance.
- Analytical support was not available as detection of the concentrations tested was not possible with any of the analytical methods available. Hence, no sampling for analysis was performed.
- The final test solutions were all clear and colourless.
- Fresh solutions of DEMB had low O2 concentration on day 3, not expected to affect test results.
- No mortality was observed in the control group.
Results with reference substance (positive control):
The 96h-LC50 was 0.16 mg/L with 95 % fiducial limits of 0.14 and 0.19 mg/L
