Description of key information

Freshwater

In a 33-day early life-stage Key test (BASF SE, 394403, 2013) with Pimephales promelas the NOEC of for growth was determined to be 0.297 mg/L based on measured concentration and the EC10 was determined to be 0.452 mg/L based on nominal concentration.

Marine water

In a 34-day early life-stage key test (BASF SE, 394404, 2015) with Cyprinodon variegatus the overall NOEC of the test substance for growth was determined to be 0.082 mg/L, based on mean measured concentration. The EC10 value was determined to be 0.071 mg/L (mean measured concentration).

Additional information

Freshwater

The chronic toxicity of the test substance to fathead minnow (Pimephales promelas) was evaluated in a 33-day early life-stage key test (BASF SE, 394403, 2015) under flow-through conditions. Embryos (< 4-hours old), larvae and juvenile fish were exposed to nominal concentrations of 0 (Control), 0.01, 0.032, 0.1, 0.32, 1.0 and 3.2 mg/L. The measured concentrations were determined to be 0.082, 0.20, 0.47, 1.1 and 2.7 mg/L. The samples of the test solutions collected during the test had measured concentrations that ranged from 81.2 to 124 % of nominal concentrations. Additional samples collected on Day 16 when the saltwater pump malfunctioned had measured concentrations ranged from 89.1 to 107 % of nominal, while the additional samples collected on Day 17 of the test when the saltwater pump resumed its normal operation had measured concentrations ranged from 81.2 to 101 % of nominal. Each test group consisted of 100 eggs evenly distributed among four replicates in glass vessels containing approx. 1.7 liters test water. The test water was charcoal filtered and diluted with deionized water. The juveniles were kept in 9-Lstainless steel aquaria. Measured and/or determined biological parameters were survival of the embryos until hatch, number of surviving larvae at end of swim-up (day 6) and survival of young fathead minnow until the end of exposure (day 33), time to hatch and swim-up, signs of toxicity; body weight (wet body weight) and total body length (from tip of the snout to the end of the caudal fin) of surviving fish were determined at the end of exposure. The biological results were based on nominal concentrations. Growth was the most sensitive endpoint. At the end of the exposure the mean wet weights and the total body lengths of the surviving fish were statistically significantly decreased at concentrations of 1.0 and 3.2 mg/L. Exposure to the test substance over 33 days resulted in no adverse effects on the time to hatch, hatching success and survival of Pimephales promelas during early life-stage development. The NOEC was 0.32 mg/L, based on nominal concentration and 0.297 mg/L based on mean measured concentrations. EC10 values were determined to be 1.262 mg/L and 0.452 mg/L (based on nominal concentrations) for length and growth, respectively.

Marine water

The chronic toxicity of the test substance sheepshead minnow (Cyprinodon variegatus) was evaluated in a 34-day early life-stage key test (BASF SE, 394404, 2015) under flow-through conditions.

Embryos (~21 - 22 hours old), larvae and juvenile fish were exposed to nominal concentrations of 0.077, 0.19, 0.48, 1.2 and 3.0 mg/L, corresponding to mean measured concentrations of 0.082, 0.20, 0.47, 1.1 and 2.7 mg/L, respectively. The study included also a negative and a solvent control. Each test group consisted of 80 embryos evenly distributed among four replicates in cylindrical glass vessels in 9-L glass aquaria containing approx. 7 L test water. The test water was sand filtered seawater diluted with freshwater to a salinity of 20 ‰. Measured and/or determined biological parameters were survival of the embryos until hatch, number of surviving larvae at end of swim-up (day 6) and survival of young sheepshead minnow until the end of exposure (day 34), time to hatch and swim-up, signs of toxicity; body weight and total body length of surviving fish were determined at the end of exposure.

The biological results were based on mean measured concentrations. Growth and body length were the most sensitive endpoints. At the end of the exposure there were statistically significant reductions in total length and wet weight among fish in the 0.082, 0.20, 0.47, 1.1 and 2.7 mg/L, and in dry weight among fish in the 0.20, 0.47, 1.1 and 2.7 mg/L treatment groups in comparison to the pooled controls. Exposure to the test substance resulted in no adverse effects on the time to hatch or swim up, hatching success and survival of Cyprinodon variegatus during early life-stage development.

The overall NOEC of the test substance for growth was determined to be 0.082 mg/L, based on mean measured concentration. The EC10 value was determined to be 0.071 mg/L (based on mean measured concentration).
