yes
Details on sampling:
- Concentrations: 0, 0.32, 1.0, 3.2, 10, 32 mg/L as nominal concentrations based on test substance mass without correction for purity
- Samples were taken on the following days:
Interval / Week 1
Day 0 (fresh solutions)
Day 2 (fresh and 48 hour old test solution)
Day 5 (fresh and 72 hour old test solution)
Day 7 (48 hour old test solution)
Interval / Week 2
Day 12 (fresh solutions)
Day 14 (48 hours old test solutions)
Interval / Week 3
Day 19 (fresh solutions)
Day 21 (48 hours old test solutions)
- Sample storage conditions before analysis: Sample transport to the Analytical Laboratory was done on the day of sampling. Storage at room temperature. The retained samples were stored in a refrigerator until sent to the analytical laboratory.

Test solutions

Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
The test item was homogenized by shaking the container before use and all test solutions were prepared individually (differential loading). The test substance is weighed, placed in a volumetric flask and filled to the mark with test medium and closed with a glass stopper. The solution was stirred for approximately 30 min at 20 ± 2 °C and visually checked for any undissolved test substance. All stock solution appeared colorless-clear

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: Daphnia magna STRAUS
- Source: The clone of Daphnia magna STRAUS 1820 used was supplied by the Institut National de Recherche Chimique Appliquée, France, in 1978.
From this date on this clone was cultured and bred continuously in the Laboratory of Ecotoxicology of the Experimental Toxic
- Body length of parent animals: 4.4 mm before exposure
- Feeding during test: During the test daphnids were fed daily a diet of live green algae Desmodesmus subspicatus, cultured in a synthetic medium. The algae were separated from their culture medium by centrifugation, resuspended in daphnid's medium (M4) corresponding to concentrations of 1110 mg and 1160 mg TOC*/L (respectively) in the algal concentrates used. The daphnids were fed a defined volume (≤180 μL) of the concentrate to reach the amount of food defined in the table below. The algae were stored in a refrigerator (dark, about 4-8°C) for maximum 21 days.

Details on test conditions:
TEST SYSTEM
- Test vessel: Numbered glass beakers (nominal volume 50 mL), sealed with ground glass stoppers. The test vessels were sealed and completely filled due to the volatility of the test substance.

- No. of organisms per vessel: 1
- No. of vessels per concentration (replicates): 10 (10 replicates)
- No. of vessels per control (replicates): 10 (10 replicates)

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: A synthetic fresh water (Elendt M4) is used as media for culture and test purposes. For the composition of this M4 medium see OECD 211.

Details on results:
- Mortality of parent animals: 30% in the highest test group (32 mg/L) after 21 days of exposure
- Reproduction: 46% in the highest test group (32 mg/L) after 21 days of exposure
- Body length of parent animals: 4.4 mm before exposure, 4.2 mm in the highest test group (32 mg/L) after 21 days of exposure

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
A significant effect on parent mortality and reduced reproductive output was observed in the highest test group (32 mg/L) after 21 days of exposure. In addition a significant effect on parent growth (as length) was also observed in the highest test group. As effects were only observed in the highest test group, ECx point estimates could not be reliably calculated.
