Short-term toxicity to aquatic invertebrates

Experimental data, supported by a QSAR prediction, indicate that there would be no effect at the limit of solubility of the substance.

An unpublished short-term toxicity study was conducted on Daphnia magna (ABC Laboratories, 1979) which reported a 48 hr LC50 of 4.5 mg/L. The levels tested (and the resulting effect level) were greatly in excess of the water solubility of the substance. Clumping of the animals at 1.0 and 1.8 mg/L was observed and there are limitations on reporting, however this study is acceptable for use as a key study towards the conclusion that there is no short-term effect on aquatic invertebrates at the limit of solubility of the substance.

 

In support of this result a QSAR (ECOSAR, USEPA 2008) is used. This QSAR predicts that the effect level for short-term toxicity of the substance to Daphnia magna is greatly in excess of its water solubility.

A further indication that there is no toxicity from this substance at the limit of solubility is that the EU Risk Assessment Reports for each of the structurally similar substances DEHP, DIDP and DINP determine that signs of toxicity to aquatic invertebrates for these substances at concentrations below their water solubilities cannot be determined.
