In different studies, 3 Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) has been investigated for reproductive toxicity to a greater or lesser extent. Often are the studies based on in vivo experiments and estimated data in rodents, i.e. most commonly in rats for Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) along with the study available on structurally similar read across substance Ametryne (CAS no 834-12-8). The predicted data using the OECD QSAR toolbox has also been compared with the experimental studies.

National Library of Medicine (HSDB (Hazardous Substances Data Bank); US national Library of Medicine reviewed by SRC, 2017) andEnvironmental Protection Agency (NTRL (National Technical Reports Library), Environmental Protection Agency, 1988) on structurally similar read across substance Ametryne (CAS no 834-12-8),male and female rat were treated withAmetrynein the concentration of0, 100, 250 and 500 mg/kg/dayorally bygavage for 28 days. 7 animals died out of 10 at 500 mg/kg bw and 1 animals died out of 10 at 250 mg/kg bw. Decrease in body weight gain were observed in treated rats at 250 and 500 mg/kg bw. Similarly, severe vascular congestion, centrilobular liver necrosis and fatty degeneration of individual liver cells were observed in treated rats at 500 mg/kg bw. In addition, no major degenerative changes were observed in liver, kidney, spleen, pancreas, heart, lung, intestine and gonads of treated rats at 250 mg/kg bw. Therefore, NOAEL was considered to be 250 mg/kg/day for P generation when male and female rats treated withAmetryneorally bygavage for28 days.

Thus, based on the above study and predictions on Bis[[4-[[4-(diethylamino)phenyl][4-(ethylamino)-1-naphthyl]methylene]cyclohexa-2,5-dien-1-ylidene]diethylammonium] dicopper(1+) hexa(cyano-C)ferrate(4-) and its read across substances, it can be concluded that NOAEL value is 205 mg/kg bw with no effect on reproduction.
