Description of key information

The toxicity of N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine was tested according to OECD guidelines 201 and 202 for acute toxicity studies with aquatic invertebrates (Daphnia magna) and algae (Pseudokirchneriella subcapitata). No toxic effect on algae was observed up to the limit of water solubility (NOEC ≥ 100 mg/L nominal, corresponding to 220 µg/L measured). For D. magna a 48 h EC50 of 170 µg/L (meas. geom. mean) was determined. The 48 h LOEC was set at 50 µg/L.

Toxicity to invertebrates:

Daphnia magna Straus was exposed for 48 hours to N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine in a static short-term toxicity study according to the OECD guideline 202 (2004) and in compliance with the OECD-GLP principles (Schwartz 2018) at nominal concentrations of 25, 50, 100, 200, and 400 mg/L. As the water solubility was determined with 0.14 mg/L in distilled water (see section 4.8 in the present dossier), some concentrations could not be verified analytically because they were below the LOD (= 0.05 mg/L). For test solutions within which the concentrations were below the LOD, the LOD was used for determination of the concentration response curve, resulting in a 48 h EC50 of 170 µg/L. According to the Guidance on Information Requirements and Chemical Safety Assessment, Chapter R.7b: Endpoint specific guidance, Version 4.0, June 2017 (Table R.7.8-3) the LOD was set as the LOEC.

Due to analytical problems a robust dose response curve could not be derived for 48 h exposition. Nevertheless, the presented EC50 value of 0.17 mg/L is conservative (being below the concentration of observed EC30) and therefore presented in the present dossier. The data also prove that the EC50 is below 1 mg/L and useful as reliable key information for classification.

Toxicity to algae:

The freshwater algae Pseudokirchneriella subcapitata was exposed to N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]- 2'-deoxyadenosine for 72 hours in a static toxicity study according to the OECD guideline 201 (2006) and in compliance with the OECD-GLP principles. The study was conducted as limit test with a measured concentration of 220 µg/L (limit of water solubility; nominal concentration of 100 mg/L). The test concentrations were determined analytically by HPLC-UV. No effect on P. subcapitata was observed.
