Two study reports (rated as Klimisch 1 and 2) on aquatic organisms are available covering two trophic levels: a primary producer (freshwater algae, represented by the model species Pseudokirchneriella subcapitata) and a primary consumer (aquatic invertebrates, represented by the model species Daphnia magna).

P. subcapitata was exposed for 72 hours to N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine in a growth inhibition test (according to OECD guideline 201 and under GLP) at a limit concentration of 220 µg/L (measured, limit of water solubility, loading 100 mg/L nominal). The test concentration was determined analytically by HPLC-UV. No effect on P. subcapitata was observed. Therefore, N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine is not toxic to P. subcapitata up to the limit of water solubility.

The aquatic invertebrate D. magna was exposed to N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine for 48 hours in a GLP toxicity study (OECD 202) at nominal concentrations of 25, 50, 100, 200, and 400 mg/L. As the substance is poorly soluble (water solubility of 140 µg/L in distilled water, see section 4.8 in the present dossier) only low concentrations were achieved after shaking the test solution for 48 h. Since a LOQ for the analytical method of less than 1.0 mg/L could not be achieved, none of the test concentrations could be verified analytically with sufficient reliability. Nevertheless, test substance concentrations could be measured in the range above the LOD (= 0.05 mg/L) but below the LOQ in the two highest treatment levels (200 and 400 mg/L nominal). Based on immobilization rates and measured concentrations, an EC50 (48 h) of 170 µg/L could be determined. 

Since N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine is not readily biodegradable, and the lowest effect value was < 1 mg/L (EC50 = 170 µg/L, LOEC ≥ 50 µg/L), N-Benzoyl-5'-O-[bis(4-methoxyphenyl)phenylmethyl]-2'-deoxyadenosine has to be classified as "Aquatic Acute 1" (H400) and “Aquatic Chronic 1 (H410)” following the provisions laid down in the CLP Regulation (Regulation (EC) No 1272/2008).
