Only one study is available and is considered as a key study (Guillot, 1981). In a dermal irritation study performed similarly to the OECD test guideline No. 404 (Draize test), 4 New Zealand White rabbits were exposed to 0.5 mL of undiluted 2 -methyl-3 butenenitrile. Test sites were covered with an occlusive dressing for 24 hours and weren’t washed after this period. Changes were observed at 25 and 72 hours. Mean score were 0.0 for erythema and edema.

 

Eye irritation

Only one study is available and considered as a key study (Guillot, 1981). In an eye irritation study performed similarly to the OECD test guideline No. 405, 0.1 mL of undiluted 2 -methyl-3 -butenenitrile(2M3BN) was instilled into the conjunctival sac of left eye of 4 New Zealand White rabbits. The eyes of the animals were not rinsed after instillation. Changes were observed at 1, 24, 48, 72, 96 and 168 hours after dosing and grading according to Draize scale. 2M3BN produced chemosis (mean score 0.5),conjunctival redness (mean score 0.5), and iridial lesions (mean scores 0.33) All these effects were completely reversed after 4 days. No corneal lesions (mean score 0.0) were observed.

Justification for classification or non-classification

Based on key studies (cited above), a self classification is proposed:

    

- As no signs of irritation were observed in a dermal irritation study with 2 -methyl-3 -butenenitrile (all mean scores = 0.0) conducted similarly to OECD test guideline No. 404, 2 -methyl-3 -butenenitrile is not classified as irritating to the skin according to the CLP Regulation (1272/2008) and the Directive 67/548/EC.

- 2 -methyle-3 -butenenitrile is slighlty irritating to eyes in a study conducted similarly to the OECD test guideline No. 405. However, no classification is required according to the CLP Regulation (1272/2008) and the Directive 67/548/EC.
