In different studies,2-amino-5-[(4-amino-3-sulfonatophenyl)(3-sulfocyclohexa-2,5-dien-1-ylidene)methyl]-3-methylbenzenesulfonate (3244-88-0)has been investigated for acute oral toxicity to a greater or lesser extent. Often are the studies based on in vivo experiments and estimated data in rodents, i.e. most commonly in rats for2-amino-5-[(4-amino-3-sulfonatophenyl)(3-sulfocyclohexa-2,5-dien-1-ylidene)methyl]-3-methylbenzenesulfonate (3244-88-0). The predicted data using the OECD QSAR toolbox has also been compared with the experimental studies.

Also these results are further supported by the experimental study conducted by Greim H, J. Ahlers et.al. (Chemophere. Vol. 28, No. 12, pp. 2203.2236, 1994) for the structurally similar read across substance 6-aminonaphthalene-1,3-disulfonic acid (118-33-2), In acute oral toxicity study, rat were treated with 6-aminonaphthalene-1,3-disulfonic acid in the concentration of 2000 mg/kg bw orally.  50% mortality was observed in treated rat at 2000 mg/kg bw. Therefore LD50 was considered to 2000 mg/kg bw when rat were treated with 6-aminonaphthalene-1,3-disulfonic acid orally.  

Also these results are further supported by the experimental study conducted by U.S. National Library of Medicine (ChemIDplusA TOXNET Database, 2017) for the structurally similar read across substance 2-aminonaphthalene-1,5-disulfonic acid (117 -62 -4).In a acute oral toxicity study, rat were treated with 2-aminonaphthalene-1,5-disulfonic acid (117 -62 -4) orally. 50% mortality was observed in treated rat at 5430 mg/kg bw. Therefore, LD50 was considered to be 5430 mg/kg bw when rat were treated with2-aminonaphthalene-1,5-disulfonic acid orally.    

Thus, based on the above studies and predictions on 2-amino-5-[(4-amino-3-sulfonatophenyl)(3-sulfocyclohexa-2,5-dien-1-ylidene)methyl]-3-methylbenzenesulfonate (3244-88-0)and its read across substances, it can be concluded that LD50 value was 4137.6 mg/kg bw. Thus, comparing this value with the criteria of CLP2-amino-5-[(4-amino-3-sulfonatophenyl)(3-sulfocyclohexa-2,5-dien-1-ylidene)methyl]-3-methylbenzenesulfonate (3244-88-0)can be “Not classified” for acute oral toxicity.
