Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
- Concentrations: algal medium (0 mg/L), 100 mg/L nominal

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method:
* preparation of stock solution: 110 mg of 3-chloro-1,2-propanediol (alphaMCG) in 1 liter algal medium resulting in a concentration of 0.11 g/L
* test solution: 90 mL of stock solution and 10 mL of algal medium resulting in a concentration of 100 mg/L

Test organisms

Test organisms (species):
Raphidocelis subcapitata (previous names: Pseudokirchneriella subcapitata, Selenastrum capricornutum)
Details on test organisms:
TEST ORGANISM
- Common name: freshwater green alga
- Strain: ATCC 22662
- Source (laboratory, culture collection): Culture collection of algae and protozoa, Ambleside, Cumbria, UK
- Method of cultivation: 100-mL Erlenmeyer flasks with 50 mL filter sterilised medium were used for algae culture. Each week 0.5-2 ml of the culture is transferred to fresh algal medium. The preculture used for the study was 4 days of age at the start of the test.
