The test item was tested for toxicity towards Daphnia magna STRAUS at different concentrations
(aqueous eluates/”WAF”2) from different amounts of the poorly water soluble test item) dissolved in
medium M4 acc. Elendt, as physiological conditions for the daphnids are necessary after the
preparation of the test solutions.

The in-house cultivation of the daphnids is performed so that the animals are transferred into new
test medium (see below) at an interval of 1 to 3 days followed by feeding with Desmodesmus
subspicatus- and/or Chlorella sp.cells. Daphnids used for the test were less than 24 hours old. In
order to ensure the correct age, daphnids are transferred into fresh medium one day before the
study start in order to ensure the test animals were less than 24 hours old. Daphnids were not fed
during the study.

Based on this, the reported effect concentrations (EL values) should refer to nominal
concentrations of the test item (as “WAF”).
Details on test conditions:
The test item was applied as follows: The test item was tested for toxicity towards daphnia as aqueous extracts (“WAF”) from up to 1000 mg test item/L (final nominal concentration in the main test). For this purpose, different amounts of the test item were dispersed
in dilution water. The elution of the water soluble part (WAF) of the test item took place on a shaking machine at 130 rpm speed over 18 hr3) at room
temperature, respectively. During that time an equilibration between the test item and the water was considered to be achieved. Thereafter, the
suspensions were filtered through glass fibre filters and membrane filters in order to eliminate nondissolved parts of the test item. The eluates were
used for the test without further dilution steps.
Four parallels were prepared for the test treatment with the test item and for the control without anytest item, respectively, using 5 animals each
parallel (main test). Two parallels were prepared for each concentration and the control without any test item, respectively, using 5 animals each
parallel in the screening test.

Under the conditions used for the test, no immobilisation of the daphnids was observed at concentrations representing the water
soluble part (hereafter abbreviated “WAF”, water accommodated fraction) of the test item at
concentrations up to 1000 mg/L (only screening-test). The main test was performed as a limit test using the WAF from 1000 mg/L “308 AQN”.
Results with reference substance (positive control):
The value of the EL50 (24 hour) in the period of the present test was 0.81 mg/L potassium dichromate. This value is slightly lower than the range
of 0.9-1.9 indicated in the study plan but fully in accordance with the recommendations given in OECD Guideline 202 (Daphnia sp., Acute
Immobilisation Test) being in the range 0.6 – 2.1 mg/L.

Any other information on results incl. tables

In the control without any test item no immobilisation of the animals was observed. In the test solutions the oxygen concentration did not drop below a value of 60% of the saturation value (» 5 mg O2/L).

Executive summary:

“308 AQN” was tested for acute toxicity towards daphnia according to OECD-Test Guideline 202 (version dated 13-Apr-2004). In order to investigate the influence of the test item towards the daphnids, the swimming behaviour of the animals was recorded. Under the conditions used for the test, no immobilisation of the daphnids was observed at concentrations representing the water soluble part (hereafter abbreviated “WAF”, water accommodated fraction) of the test item at concentrations up to 1000 mg/L (only screening-test). The main test was performed as a limit test using the WAF from 1000 mg/L “308 AQN”. The following EL-values (effective loading rate) with “308 AQN” are assumed according to the results of this test (definitive test): EL50 >1000 mg/L; NOEL ≥1000 mg/L.
