Determination of the test item
All concentration levels and the control were analytically verified via HPLC-DAD at the start (0 hours) and at the end of the exposure (72 hours) with algae.

Stock solution, media preparation
100 mg main component/L, corresponding to 124.4 mg test item/L were freshly prepared with dilution water. The dispersion was agitated until the stock solution was clear.

Five concentration levels were tested in a geometrical series with a dilution factor of 2: 6.25 - 12.5 - 25.0 - 50.0 - 100 mg main component/L, corresponding to 7.77 - 15.5 - 31.1 - 62.2 - 124.4 mg test item/L. The concentration levels are based on the results of a preliminary range finding test.

Six replicates (without test item) were tested under the same test conditions as the test item replicates.

Cell density was measured daily via Chlorophyll a fluorescence, excitation at 436 nm, emission at 685 nm. Dilution water was used as a background signal. Self-fluorescence was observed in the preliminary range finding test at the concentration of 100 mg test item/L. A separate replicate of each test item concentration was prepared without algae and treated like the other test replicates. The self-fluorescence of each measuring point was taken into account in the evaluation.

The toxicity of potassium dichromate (SIGMA ALDRICH, batch number MKBV0900V, purity 99.0%, CAS RN 7778-50-9) to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined over a period of 72 hours from 2017-04-18 to 2017-04-21 (with headspace). The reference item toxicity is in the valid range following test facility SOPS.

EC10-, EC20- and EC50-values with confidence intervals of yield inhibition after 72 hours were calculated by sigmoidal dose-response regression.

NOEC, LOEC and analyses
The NOEC / LOEC was determined by calculation of statistical significance of growth rate and yield.

Shapiro-Wilk’s test on normal distribution was done with a significance level of 0.01.
Levene’s test on variance homogeneity was done with a significance level of 0.01.
Monotonicity of Concentration/Response was assessed by trend analysis by contrasts (significance level: 0.05) for yield.
Williams Multiple t-test was done with a significance level of 0.05 for yield.
Welsh-t-test after Bonferroni-Holm was done with a significance level of 0.05 for growth rate.

Conclusions:
In this study, the test item was found to inhibit the growth of the freshwater green alga Pseudokirchneriella subcapitata after 72 hours with the following effect values (based on nominal main component concentrations): The EC50-values with 95% confidence intervals for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were > 100 mg/L and 47.9 (42.3 – 54.9) mg/L, respectively. The NOEC-values for inhibition of growth rate and yield after 72 hours were 25.0 and 12.5 mg/L, respectively.
All effect values given are based on nominal main component concentrations of the test item.

Executive summary:

The toxicity of the test item to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined according to the principles of OECD 201 and Council Regulation (EC) No. 266/2016/Method C.3 from 2017-09-25 to 2017-09-28 at the test facility.The aim of the study was to assess the effects on growth rate and yield over a period of 72 hours.

 

The study was conducted under static conditions with an initial cell density of 6839 cells/mL. A stock solution with a nominal loading of 124.4 mg test item/L, corresponding to 100 mg main component/L was tested. Five concentrations were tested in a geometrical series with a dilution factor of 2 (nominal):6.25 - 12.5 - 25.0 - 50.0 - 100 mg main component/L, corresponding to 7.77 – 15.5 – 31.1 – 62.2 – 124.4 mg test item/L. Three replicates were tested for each test item concentration and six replicates for the control. The environmental conditions were within the acceptable limits.

 

The concentration of the test item Sanodure Fast Gold L dried was analytically verified via HPLC-DAD at the start of the exposure (0 hours) and at the end of the exposure (72 hours) in all tested concentrations and the control.

The measured concentration of the test item at the start of the exposure (0 hours) were between 96 and 100% of the nominal values. The measured concentrations at the end of the exposure (72 hours) were between 89 and 104% of the nominal values.

 

All effect values were based on the nominal test item and main component concentration of the test item. The results are summarized in the following table:
