PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A stock solution with a nominal concentration of 5.0 mg/L was prepared with the test item and algal growth medium (OECD Medium). The test solutions were prepared by the appropriate diluting of this stock solution and distributed into test vessels prior to introduction of algae.
- Controls: A concurrent untreated control was examined in parallel to the test item concentrations. A reference control with potassium dichromate is performed at least twice a year to demonstrate satisfactory test conditions.

- Source (laboratory, culture collection): The algae were supplied by the SAG: Collection of Algal Cultures, Inst. Plant Physiology, and University of Göttingen, GERMANY.
- Age of inoculum (at test initiation): 3 days
- Method of cultivation: Cultured under standardised conditions (OECD 201). Stock cultures were small algal colonies that are inoculated onto agar regularly. These were transferred to fresh agar medium at least once every two months and were maintained under standardised conditions according to the test guidelines.The pre-culture was intended to give a quantity of algae suitable for the inoculation of test cultures. The pre-culture was prepared with the OECD algal growth medium, incubated under the same
conditions as the test and used when still growing exponentially, normally after an incubation period of about three days.

Reported statistics and error estimates:
- The section-by-section specific growth rates in the control cultures were assessed (calculated as the specific growth rates for each day during the course of the test (days 0-1, 1-2 and 2-3) and to demonstrate exponential growth for the entire study period. The inhibition of alga growth was determined from the biomass (area under the growth curves, A), the average specific growth rate (r) and from the yield (y). Mean values and standard deviations were calculated for each concentration at the start, and at the end of the test using Excel 2007 for Windows software (Microsoft Co./One Microsoft Way/Redmond, WA 98052-6399).
- Statistical comparisons of biomass, average specific growth rates and yield in controls and in the treated groups were carried out using analysis of variance (ANOVA) and Bonferroni t-Test (a= 0.05) by TOXSTAT software. For the determination of the LOEC and NOEC, the calculated mean biomass, growth rates and yield at the test concentrations were tested on significant differences to the control values by Bonferroni t-Test.

The 72h EC50 of the test item on freshwater algae is 0.401 mg/L, the NOEC is 0.092 mg/L and the LOEC is 0.190 mg/L, based on growth rate (r).
Executive summary:

A freshwater algae growth inhibition test was performed on the test item, according to OECD TG 201, under GLP conditions. Based on a rage-finding study where significant inhibition was observed, the test was performed at concentrations of 0.051, 0.128, 0.32, 0.8, 2.0 and 5.0 mg/L test item (nominal) on Pseudokirchneriella subcapitata, alongside concurrent negative controls. Four replicates were performed per concentration and seven for the control. Growth was monitored daily by measuring the number of cells in each culture. Analytical monitoring was performed using a validated method at the start and at the end of the experiment. As the measured values differed more than 20% from the nominal values, the measured concentration values were used to calculate the results. These correspond to 0.031, 0.092, 0.190, 0.420; 1.005 and 2.502 mg/Ltest item (measured).

With respect to the inhibitory effect of the test item, the 0-72 h average specific growth rates were significantly different from that of the control group in the examined concentration range of 0.190 – 2.502 mg/L (measured), therefore the the NOEC was determined as 0.092 mg/L (measured) and LOEC was determined as 0.190 mg/L (measured), based on growth rate. The 0-72 h areas and yield were significantly different from that of the control group in the examined concentration range of 0.092 – 2.502 mg/L (measured). The NOEC was determined as 0.031 mg/L (measured); the LOEC was determined as 0.092 mg/L (measured). The ErC50, EbC50 and EyC50 values of the test item could not be calculated, due to the slight effect observed, and therefore, they were determined directlyfrom the raw data. The 72h EC50 of the test item was 0.401 mg/L, based on growth rate.
