Description of key information

Key data were identified for skin irritation. Based on assessment of the available data, dodecane-1-thiol should be considered corrosive to the skin and to the eyes. There was no evidence of respiratory irritation in rats exposed to dodecane-1-thiol for four weeks. The key parameter values are presented below:

Key data was available to evaluate the skin irritation potential of dodecane-1-thiol.

 

In a key guideline dermal irritation/corrosion study (Gerbeix, 2010; Klimisch score = 1), a single male New Zealand white rabbit was dermally exposed to 0.5 mL of undiluted dodecane-1-thiol (CAS Number 112-55-0) for 3 minutes, 1 hour, and 4 hours. Since the test material was deemed a severe irritant but not corrosive during the 4 -hour exposure on this first animal, a second animal was exposed to the test material for 4 hours. Animals were observed for 14 days and erythema and oedema scores were recorded at daily and at 24, 48, and 72 hours. The 3-minute exposure resulted in slight to severe erythema and slight or moderate oedema for the length of the observation period. The 1-hour exposure resulted in well-defined to severe erythema and slight or moderate oedema throughout the length of the observation period, in addition to scabs by day 13. Both animals exhibited well-defined, moderate, or severe erythema while the second animal also exhibited slight to severe oedema. Due to the severe cutaneous reactions observed, the second animal was sacrificed on day 8. The 4-hour exposure mean scores over 24, 48 and 72 hours for each animal were 2.3 and 3.0 for erythema and 0.0 and 1.0 for oedema. None of the symptoms noted for any exposure period were reversible within 14 days. Based on the severe erythema observed under the conditions of this study, dodecane-1-thiol is considered to be corrosive to the skin of rabbits.

Dodecane-1-thiol is classified as corrosive Category 1C (Danger; H314: Causes severe skin burns) under the EU CLP regulation and R34 (C; Causes burns) under the EU DSD regulation. Dodecane-1-thiol is classified as Category 1 (Danger; H318: Causes serious eye damage) under the EU CLP regulation.
