Reference
Endpoint:
short-term toxicity to fish
Type of information:
(Q)SAR
Adequacy of study:
key study
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
other: Calculation of toxicity using constituent data generated by EcoWin v1.00 QSAR. methodology according to EFEO NCS protocol.
Justification for type of information:
QSAR prediction: migrated from IUCLID 5.6
Qualifier:
according to guideline
Guideline:
other: NCS protocol
Deviations:
no
Principles of method if other than guideline:
NCSs being complex mixtures that are often very poorly water soluble and highly volatile, create special difficulties with the present test protocols which will require adaptation. Instead, the ecotoxicity was estimated using a QSAR and by read across from the constituents.

The combined toxicity of each NCS is calculated based on the ecotoxicity of the constituents according to the additivity formula in line with the NCS protocol (see attached file "formula.doc").

Calculated data for the constituents are obtained from the QSAR EcoWin v1.00.

Estimated ecotoxicity values of Cornmint oil constituents towards fish in mg/l.

(When more than one Ecosar class wass considered justified, the class resulting in the lowest toxicity value was used).

Substance CAS Ecosar class Fish 96hLC50 (est) in mg/l
L-menthol 2216-51-5 Neutral organics 7.336
Menthone 89-80-5 Neutral organics 20.408
L-limonene 5989-54-8 Neutral organics 0.341
beta-Pinene 127-91-3 Neutral organics 0.903
1,8-cineole 470-82-6 Neutral organics 11.968
alpha-Pinene 7785-70-8 Neutral organics 1.059
Iso-menthone 491-07-6 Neutral organics 20.408
Neo-menthol 2216-52-6 Neutral organics 7.336
Methyl acetate 16409-45-3 esters / Neutral organic SAR (baseline toxicity) 1.165
Piperitone 89-81-6 Vinyl/allyl ketones / Neutral organic SAR (baseline toxicity) 14.493
Pulegone 89-82-7 Vinyl/allyl ketones / Neutral organic SAR (baseline toxicity) 11.257
Iso-Pulegol 89-79-2 Neutral organics 7.34
Germacrene-D 37839-63-7 Neutral organics Substance outside applicability domain of QSAR; no toxicity at saturation expected
Neo-iso-pulegol 89-79-2 Neutral organics 7.34
Octan-3-ol 589-98-0 Neutral organics 22.783
Neo-iso-menthol 89-78-1 Neutral organics 7.336
Beta-caryophyllene 87-44-5 Neutral organics Substance outside applicability domain of QSAR; no toxicity at saturation expected
 Sabinene  3387-41-5  Neutral organics 0.452
 Myrcene  123-35-3  Neutral organics  0.309

The log Kow values for Germacene-D and beta-Caryophyllene lie outside the applicability domain of the QSAR. Therefore, as no toxicity is predicted by the QSAR at saturation, these substances are left out of the calculation of the toxicity of the mixture.

Validity criteria fulfilled:
not applicable
Conclusions:
The acute toxicity (96h-LC50) of Cornmint oil towards fish is 3.01 mg/l.
Executive summary:

The acute toxicity of Cornmintoil towards fish was estimated using the QSAR EcoWin v1.00 according to the NCS protocol. 96h-LC50 values were calculated for individual constituents using QSARs for one or more relevant chemical groups to which the respective constituent belongs. The worst case LC50 values (from different QSARs) for each for the constituents were used to calculate the LC50 of Cornmint oil according to the formula given in the NCS protocol (see attached file "formula.doc").

The 96h-LC50 of Cornmint oil towards fish was calculated to be 3.01 mg/l.
