Description of key information

The 72h-ErC50 of > 10.2 mg/L and the 72h-ErC10 of 5.13 mg/L were determined in a test according to OECD TG 201 (GLP, reliability 1). These values are based on measured active ingredient concentrations.

Additional information

The toxicity of nonylphenol ethoxylate, sarcosine derivative to the unicellular freshwater green alga Desmodesmus subspicatus was determined according to the principles of OECD 201 under GLP conditions. The study was assigned reliability 1 as all validity criteria were fulfilled. The aim of the study was to assess the effects on growth rate and yield over a period of 72 hours.

The study was conducted under static conditions with an initial cell density of 4332 cells/mL. Based on a preliminary test 6 concentration levels were tested in a geometrical series with a dilution factor of 2 (nominal): 0.3125 - 0.625 - 1.25 - 2.50 - 5.00 - 10.0 mg/L corresponding to following geometric mean measured concentration levels: 0.313 - 0.665 - 1.34 - 2.90 - 6.63 - 10.9 mg/L. Three replicates were tested for each test item concentration and six replicates for the control. Environmental conditions were determined to be within the acceptable limits. The concentrations of nonylphenol ethoxylate, sarcosine derivative were analytically verified by LC-MS/MS at test start and test end of all test concentrations and the control. The measured concentrations of nonylphenol ethoxylate, sarcosine derivative at test start were in the range of 109 to 127 % and after 72 hours in the range of 85 to 139 % of the nominal values. Therefore, all effect values given are based on the geometric mean measured concentrations of the test item nonylphenol ethoxylate, sarcosine derivative converted to active ingredient concentration.

After 72 hours of exposure, 0.2 to 0.5 mL aliquots of the test medium (i.e. samples of exposed algal cells) from the two highest concentrations (nominal test item concentrations 10.0 and 5.00 mg/L) and the control were transferred to 10 mL untreated test medium (three replicates for test item concentrations and duplicate for control). Algae were then allowed to grow for further 4 days under test conditions. The test item effect was observed to be reversible at these concentrations. Therefore, there is potential for recovery following exposure up to the nominal test item concentration of 10.0 mg/L (highest test concentration).
