Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Description of key information

The target substance (TS) is a multiconstituent substance, containing seven constituents. The acute daphnia toxicity prediction was made by assessing the TS as a mixture and assessingeach constituent individually first and then the mixture. Six of the seven constituents are members of the ECOSAR category "Schiff Bases-Azomethine". The seventh, benzaldehyde, the starting material for the other

constituents, is a member of the ECOSAR category “Aldehydes (mono)”.

The acute daphnia toxicity was predicted individually for each constituent, using the respective ECOSAR category as primary categorization. The specific endpoint was the 48-hour EC50 to Daphnia magna using ‘mobility’ as the effect. Six of the seven constituents, except benzaldehyde, were structurally very similar except for the carbon chain length and degree of unsaturation. Read-across for all seven constituents was done using the robust read-across methodology, integral to the OECD QSAR Toolbox.

The endpoint value for the TS (mixture) was then assessed both by ‘independent’ and ‘similar’ modes of action. The predicted value is a 48-hour EC50 of 19-20 mg/L to Daphnia magna. The lower value

of 19 mg/L is accepted for this endpoint. This is within the range of 48 -hour EC50 to Daphnia magna, for the individual constituents (14.7 - 33.0 mg/L). This is a reliable prediction and satisfies the endpoint adequately and can be used for risk assessment.

The structures and individual data points used for read-across are included in the attached data

matrix.

Additional information

The information requirement for this endpoint has been adapted according to REACH Annex XI, 1.5, read across and grouping. Also, ECHA Practical Guide 6, How to Report Read-Across and Categories, has been followed.

The predicted value for the 48-hour EC50 to Daphnia magna for the target substance (19 mg/L) has been derived by read-across from individual constituents using the OECD QSAR Toolbox. The prediction report as well as the data matrix are attached to the summary.
