The concentration of V723 in the test solutions was measured at the start (excess solution) and end of the test. At the end of the test one replicate of each test concentration was sampled.

The test organism was the freshwater crustacean Daphnia magna Straus, obtained from continuous laboratory cultures.
The stock cultures of Daphnia were maintained in a reconstituted water medium, identical to the test dilution water, at a temperature of 20 ± 2°C, and a photoperiod of 16 hours light: 8 hours dark.
The cultures were fed a defined diet of algae Chlorella vulgaris and yeast.
Culture conditions were such that the Daphnia reproduction was by diploid parthenogenesis.
Daphnia <24 hours old were used for testing.

A solution of 112 mg V723/litre was prepared by addition of V723 to the dilution medium. Information supplied with the test sample V723 indicated that it contained 10.5% of water and test concentrations were adjusted accordingly to account for a nominal test concentration of 100 mg/l.
Details on test conditions:
Borosilicate glass beakers were used as test vessels, with four replicates per exposure concentration, each containing 200 ml of test solution.
When the test solutions were at test temperature, five Daphnia were added to each vessel in <2.0 ml of dilution water, giving a total of 20 Daphnia per concentration.
The temperature was maintained at 20 ± 1°C and a photoperiod of 16 hours light: 8 hours dark was provided.
The Daphnia were not fed during the course of the test.

The 24 and 48 hour median effective concentrations of Substance HI 12339 to the freshwater crustacean Daphnia magna (<24 hours old) were determined under static test conditions at a temperature of 20 ± 1°C.
The median effective concentration (EC50) is defined as the concentration, calculated from the data obtained, resulting in immobilisation of 50% of the Daphnia in the time period specified.
