Description of key information

Isopropylidenglycerinacrylate is rather water soluble (app. 35g/L) with a corresponding log Pow of 1.4. In combination with its low molecular weight of 186.2g/mol, oral and dermal absorption is expected. Dermal uptake might also be enhanced by its irritant properties that result in damage to the epithelial barrier and was confirmed by the positive results obtained in an in vivo skin sensitization assay.

Based on the log Pow value between -1 and 4, respiratory absorption is likely, but considered less relevant due to the low vapour pressure.

Metabolism has been predicted with the help of OASIS Times. After absorption, the substance will probably be quickly metabolised by esterases to the corresponding alcohol (2,2-dimethyl-1,3-dioxolan-4-ylmethanol, CAS 100-79-8) and acrylic acid. Acrylic acid in turn will be beta-oxidized and exhaled as CO2. The remaining alcolhol will most likely be conjugated, e.g., to glutathion, and renally excreted. No accumulation is expected.

No systemic toxicity has been observed for the parent compound that clearly was not secondary to local irritation, which was also the case for the proposed metabolites.
