PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: The test solutions were prepared by dilution of a stock solution. A stock solution of test item at a nominal concentration of 50 mg/L was prepared with OECD alga growth medium. 0.0250 g of test item was weighed and dissolved in 500 mL of growth medium by magnetic stirring for about 5 hours. After the test item was dissolved completely, the solution was used to dilute test solutions. Pipette 1.0, 3.2, 10, 32, and 100 mL of the stock solution and make up to 500 mL with OECD alga growth medium. The solutions were mixed thoroughly, resulting in test solutions at 0.10, 0.32, 1.0, 3.2 and 10 mg/L.

Physicochemical Measurements in the Definitive Test:
During the test, the incubation temperature was in the range of 22.3 ~ 23.8 oC. The measured light intensity was 5290 ~ 5450 lux for cool white light at the beginning and the end of the test. The light intensity was in the range of -1.45% ~ 1.53% from the average light intensity, i.e. 5368 lux over the incubation area. The values of pH determined at the beginning and the end of the test were within the range of 7.94 - 8.32. The pH of the control medium increased by 0.15 units maximally during the test.

Results of test culture in the Definitive Test:
After the 72-hour exposure period, the mean growth yield of control cultures was 4.57E+05 cell/mL, and the cell density of control cultures increased by a factor of 91.4 in the control replicates. The mean coefficient of variation for section-by-section specific growth rate in control cultures was 28.5%. The coefficient of variation of average specific growth rates during the whole test period in replicate control cultures was 0.4%. The results met the requirements of the test validity.
During the 72-hour test duration, there was no abnormal appearance of the algae observed in control replicates and replicates of each test group. After 72 hours exposure, comparing with the control group, the test groups showed increasing inhibition effects with increasing concentration. There was no significant difference in algae growth rate and growth yield between the control group and the nominal concentration of 0.10 mg/L test group (p>0.05, t-test).

Verification of Test Concentration in the Definitive Test:
The concentrations of test item in the test media were determined by HPLC method. Analysis of the concentration of the test item were performed in 0 hour fresh solutions and 72 hours old solutions. The results showed that the determined concentrations in 0-hour fresh media were 96.3% ~ 113.1% of nominal concentrations, the determined concentrations in 72 hours old media were 83.0% ~ 112.8% of nominal concentrations. The test concentrations were satisfactorily maintained in the range of 80 - 120% of nominal concentration.
