DOSAGE PREPARATION:
FAT 31016/C was suspended in polyethylene glycol to achieve the corresponding dosage level. Before treatment the suspension was homogeneously dispersed with an Ultra-Turrax and during treatment it was kept stable with a magnetic stirrer.

Fasting details:
Animals fasted overnight were treated by single oral intubation. Physical condition and rate of deaths were monitored throughout the whole observation period.

Executive summary:

The acute oral LD50 of FAT 31016/I was determined in a study conducted according to OECD Guideline 401. Groups of 5 males and 5 females each, were administered the test substance at the doses of 1000, 2500 and 5000 mg/kg bw. Clinical signs and mortality check were recorded during an observation period of 14 days. Body weights were recorded immediately prior to dosing (control weights) and at 7 and 14 days. The treated rats were submitted to necropsy whenever they died, survivors at the end of the observation period. One male was found dead in animals treated at 1000 mg/kg bw. 1 male and 2 females at the dose of 2500 mg/kg bw, while 5 males and 5 females at the dose of 5000 mg/kg bw were found dead during the observation period. Acute inhalation toxicity

Currently, acute inhalation toxicity study with Basic Red 046 Bromide is not available. However, it can be considered to be not available for inhalation owing to the low volatility on account of high melting as well as boiling point, and high water solubility of 7.9 g/L, indicates if vapours are produced will be trapped in the mucus. Further, Basic Red 046 Bromide was found to have low acute oral toxicity potential (LD50 = 2289 mg/kg bw), which supports the conclusion that no more severe effects than already seen with acute oral exposure, are expected via inhalation route. Taking into consideration the above discussion, further experiments to assess acute inhalation toxicity were considered scientifically not necessary.

Acute dermal toxicity:

Currently no study to evaluate the acute dermal toxicity of Basic Red 046 Bromide is available. The molecular weight of Basic Red 046 Bromide is 401.3 g/mol, which indicates substance is too large for dermal absorption. The high water solubility (7.9 g/L) and low partition coefficient (log Pow = -1.43) of Basic Red 046 Bromide, indicate the substance may be too hydrophilic to cross the lipid rich environment of the stratum corneum. Hence, the dermal uptake for the substance is expected to be low.

Basic Red 046 Bromide was found to have low acute oral toxicity potential (LD50 = 2289 mg/kg bw). Further, the absence of systemic toxicity or mortality in skin irritation as well as sensitization studies, further supports the conclusion that no more severe effects than already seen with acute oral exposure, are expected via dermal route. Taking into consideration the above discussion, further experiments to assess acute dermal toxicity were considered scientifically not necessary.

Based on the available information, Basic Red 046 Bromide does not warrant classification for acute toxicity as per the Regulation EC No. 1272/2008 (CLP) criteria.
