Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
- Concentrations: 100 mg/L (nominal)
- Sampling method: Two sets were collected (fresh and old medium at sample preparation, renewal and the end of exposure). Samples were taken form the middle layer of the relevant vessels.

Test solutions

Vehicle:
no
Details on test solutions:
Test solutions were prepared as Water Accommodated Fractions (WAFs) and Water Soluble Fractions (WSFs). Initially amounts of test substance were weighed and added directly to aspirators where they were stirred for 48 hours. The resulting WAF was then filtered using a 0.45 µm membrane filter. The resulting solutions were then used on study. This procedure was confirmed as a result of the range finding phase, where the physical presence of the test material caused immobility in Daphnids.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: Daphnia magna
- Source: In house cultures
- Feeding during test: No

ACCLIMATION
- Acclimation period: 28 days or less
- Acclimation conditions (same as test or not): same as test
- Type and amount of food: Chlorella vulgaris 6 mgC (organic carbon) /2 L/day
- Feeding frequency: As above
- Health during acclimation (any mortality observed): None observed

METHOD FOR PREPARATION AND COLLECTION OF EARLY INSTARS OR OTHER LIFE STAGES: Juveniles less than 24 hours old were used for the test.

Remarks on result:
other: exceeds limit of aqueous solubility
Details on results:
No immobility was observed at a limit concentration of 100 mg/L (loading rate). This mean test concentration at this loading rate was 0.00970 mg/L. The test solutions used on study were colourless with no precipitate evident and the effect concentrations exceeded the solubility of the test substance.
Results with reference substance (positive control):
- Results with reference substance valid: Yes (within historical laboratory range)
Reported statistics and error estimates:

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Remarks:
validity criteria on test fulfilled
Conclusions:
The effect concentrations to Daphnia magna exceeded the aqueous solubility of the test substance when performing a limit test at a loading rate of 100 mg/L. The water solubility of the test substance in the aqueous media was determined to be very low.
