All acute toxicity test were not performed with Tert-amyl hydroperoxide, however, missing data are available by Read-Across with the analogue substance 1,1,3,3-Tetramethylbutyl hydroperoxide.

 

Aquatic invertebrates

 

The acute toxicity test to Daphnia magna was not performed with Tert-amyl hydroperoxide, but through analogy with 1,1,3,3-Tetramethylbutyl hydroperoxide, the data obtained is considered for the registered substance: an acute toxicity test to Daphnia magna was conducted in accordance with OECD test guidelines and with the OECD Principles of Good Laboratory Practice. The toxicity of the test chemical and any formed degradation products to juvenile daphnia was determined in a static system over an exposure period of 48 hours: the NOEC was determined as 4.21 mg/L (Measured Mean Concentration) ;the EC50 (48h) was calculated as 6.7 mg/l. (Measured Mean).

 

 

Algae

 

Regarding toxicity to algae, 2 studies are available. The first one was performed with the registered substance and the second, which is used as supporting study, with the analogue substance 1,1,3,3-Tetramethylbutyl hydro peroxide.

The determination of the growth inhibition of the freshwater algae Pseudokirchneriella subcapitata exposed to the test item TERT-AMYL PEROXIDE for duration of 72 hours was assessed according to the OECD Guideline 201 in a GLP study. The results were as follows (extrapolated concentrations as geometric mean between measured initial and final abiotic values):

 

ErC50-72h = 1.2 mg/L

ErC10-72h = 0.38 mg/L

 

In order to predict the effects of 1,1,3,3-Tetramethylbutyl hydro peroxide in an aquatic environment, an Algal Growth Inhibition test was conducted in accordance with OECD test guidelines and with the OECD Principles of Good Laboratory Practice.  

 

The toxicity of the test chemical to an exponentially growing culture of P. subcapitata was determined in a sealed system over an exposure period of 72 hours: the ErC10(72h) was calculated as 2.9 mg/L (Growth rate). The ErC50(72h) was calculated as 5.6 mg/L. (Growth rate Geometric Measured Means).

 

These tests on algae indicate that both substances are "Toxic" to algae.

 

Microorganisms

 

There is no test performed with Tert-amyl hydroperoxide to assess the toxic effects on microorganisms. This data is read-across from data obtained with1,1,3,3-Tetramethylbutyl hydroperoxide.

The toxicity to activated sludge was determined in accordance with OECD Test Guideline 209, and in compliance with the OECD principles of Good Laboratory Practice: the EC50 of 1,1,3,3-tetramethylbutyl hydroperoxide for activated sludge after 3 hours contact time is 138 mg/L. The EC10 after 3 hours contact time is 33 mg/L.

 

Fish

 

Referring to the position paper "OP Consortium PP_Acute to Chronic Ratio.pdf" (attached to IUCLID), it can be conclude that algae and daphnids are the most sensitive species when tested with organic peroxides. As a consequence, no test on fish have been performed.
