Description of key information

The EC50 value, for 2 -Propenic aicd, C12 -14-alkyl ester, was determined to be greater than 10000 mg/L and also the EC10 was determined to be greater than 10000 mg/L.

In the toxicity to microorganisms test the EC50 value of the test substance, 2-Propenoic acid, C16-18-alkyl esters determined to be greater than 1000 mg/L.

The EC50 value for 2 -Proenoic aicd, C18-26-alkyl ester was determined to be greater than 1000 mg/L and also the EC10 was determined to be greater than 1000 mg/L.

Based on this result the EC50 value of 2-Propenoic acid, octadecyl ester is also considered to be >1000 mg/L.

The inhibition of the degradation activity of activated sludge, with 2 -Propylheptyl acrylate, is not anticipated when introduced in appropriate low concentrations.

Additional information

In the key study, the test substance 2 -Propenoic aicd, C12 -14 -alkyl ester was tested with Pseudomonas putida. The test concentrations were 100, 1000 and 10000 mg/L. The EC0 was >= 10000 mg/L. and the EC10 was also greater than 10000 mg/L. In the supporting study, the ciliate toxicological assessments were performed following the protocol described by Schultz (1997). The test item was not toxic towards Tetrahymena pyroformis at concentrations up to saturation of the test water with the compound .

The objective of this key study according to OECD No. 209 was to determine the effect of different concentrations of the test substance, 2-Propenoic acid, C16-18-alkyl esters on the oxygen cossumption of microbial-inoculum, compared to the blank control. The test was conducted at concentrations of 62.5, 125, 250, 500 and 1000 mg/L. Based on these results, the EC50 value was calculated to be greater than 1000 mg/L.

The EC50 value of for the reference substance 3,5-dichlorophenol was calculated to be 7.4 mg/L, which was within the acceptable limits, (5.0 to 30 mg/L) as specified in OECD 209 Guideline. Thus, all validation criteria were fulfilled.

A determination of the Inhibition of oxygen consumption in the activated sludge respiration Inhibition test for the the read across test substance 2 -Propenoic acid, C18 -26 -alkyl ester was performed. The study was conducted according to the following test guidelines: OECD Guideline 209. The test was conducted at concentrations 1000, 500, 250, 125, 62.5 mg/L as nominal concentration based on test substance without correction of purity or composition and blank controls. Incubation time was 3 hours. Sludge concentration in the test: 1.5 g/L and the inoculum was Activated sludge from a municipal wastewater treatment plant from the municipal wastewater treatment plant of Mannheim, Germany.

Based on these results, the EC50 value was calculated to be greater than 1000 mg/L.

The EC50 value of for the reference substance 3,5-dichlorophenol was calculated to be 5.3 mg/L, which was within the acceptable limits (5.0 to 30 mg/L) as specified in OECD 209 Guideline. Thus, all validation criteria were fulfilled.

In a test with domestic activated sludge the inhibition of oxygen consumption was examined according to guideline ISO 8192 (BASF AG, 1988). After 30 minutes oxygen uptake was inhibited by 7 % at 1000 mg/L, the highest concentration tested. As no reference substance was tested and therefore, the sensitivity of the activated sludge is unknown, the test result should be used with care. EC20 > 1000 mg/L

The toxicity to microorganisms, with 2 -Propylheptyl acrylate, was determined in a GLP short-term respiration test according to OECD guideline 209 using domestic activated sludge from a municipal wastewater treatment plant. The respiration rate (oxygen consumption) of an aerobic activated sludge fed with a standard amount of synthetic sewage was measured with test item concentrations of 62.4, 125, 250, 500, 1000 mg/L. Further the reference substance 3,5 - dichlorophenol and two blank controls were tested. One test assay per concentration was tested. After 180 min an EC10 of 340 mg/L and an EC50 of 1000 mg/L was determined (BASF SE, 2009).

Based on this result the EC50 value of 2-Propenoic acid, tetradecyl ester is also considered to be >1000 mg/L.
