PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances):
Preparation of stock solution (0.5 mg/mL): -> Stock solution (0.1 mg/mL): 10 mg of the test item was weighed into a 100 mL volumetric flask, filled up to the mark with acetonitrile and thoroughly mixed.
Eluent: -> Acetonitrile : water +0.1% Formic acid - gradient

The stock cultures are small algal cultures that are planted on agar regularly. These are transferred to fresh medium at least once every two months under standardised conditions according to the test guidelines. The pre-culture is intended to give an amount of algae suitable for the inoculation of test cultures. The pre-culture was prepared with Algal Mineral Salts Culture Medium, incubated under the conditions of the test and used when still exponentially growing, normally after an incubation period of about three days (The pre-culture was incubated for three days at this test.)- Test type:

In a pre-test nominal concentrations up to 100, in the main study nominal concentrations of 0.5; 1.5; 4.9; 15.6 and 50 % v/v saturated solution, corresponding to calculated test item concentrations (based on the analytical measurements) of 0.2; 0.6; 1.8; 5.9 and 20.1 mg/L, respectively. Reference
The ErC50, EbC50 and EyC50 values of the test item and their confidence limits were calculated using Probit analysis by TOXSTAT software. For the determination of the LOEC and NOEC, the calculated mean biomass, growth rates and yield at the test concentrations were tested on significant differences to the control values by Bonferroni t-Test. Biological results are related to the geometric mean of the measured test item concentrations during the test.
