In accordance with Guidelines for the Testing of Chemicals (HJ/T 153-2004) and Chemical Test Methods – Ecological System Effects Volume (second edition) (Beijing: China Environment Press, 2013), and with reference to Procedure 209 of the Guidelines for the Testing of Chemicals of the OECD, this project used activated sludge from a domestic sewage treatment plant in China to measure the three-hour respiration inhibition effects of the test substance on activated sludge. Activated sludge was exposed to three test substance concentrations of 10, 100, and 1000 mg/L, and the three-hour respiration rate (mg O2/L ∙ h) was measured. The activated sludge respiration inhibition rate was obtained by calculating the test substance respiration rate and the inoculum blank control respiration rate. 


The test results indicated that the test dissolved oxygen of the substance before and after the abiotic control test was 8.10 and 8.00 mg O2/L, there were no obvious abiotic aerobic effects; the 3h-EC50 value for the reference compound 3,5-dichlorophenol was 9.41 mg/L and the 95% confidence interval was 7.88-11.2 mg/L. For the two blank controls, the respiration rates were 35.9 mg O2/L ∙ h and 37.5 mg O2/L ∙ h, which converted to activated sludge (1.5 gdwt/L) respiration rates of 23.9 mg O2/g ∙ h and 25.0 mg O2/g ∙ h (> 20 mg O2/g ∙ h). For the two blank controls, the difference in respiration rates was 3.21%.


The activated sludge 3h-EC50 value for the test substance was >1000 mg/L.
