PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: Preparation of test solutions started with the highest concentration of 100 mg/L applying a one-hour period of magnetic stirring to accelerate the dissolution of the test item in test
medium. Lower test concentrations were prepared by subsequent dilutions of the highest concentration in test medium.
- Other relevant information: All test solutions were clear and colorless at the end of the preparation procedure.

TEST ORGANISM
- Common name: Carp (Cyprinus carpio, Teleostei, Cyprinidae) Linnaeus, 1758.
- Source: De Haar Vissen’, Zodiac proefacc., Wageningen University and Research Centre, The Netherlands.
- Length at study initiation: 2.8 ± 0.4 cm.
- Weight at study initiation: 0.23 ± 0.12 g.
- Characteristics: F1 from a single parent-pair bred in UV-treated water.
- Total fish used: 20.

HOLDING
- Quarantine/Acclimatisation: At least 12 days after delivery.

TEST SYSTEM
- Test vessel: Control and highest concentration: 10 litres, containing 8.0 litres of test solution. Intermediate concentrations: 3.5 litres, containing 3.0 litres of test solution.
- Test type: Static.
- Aeration: no aeration.
- Number of fish: Control and highest concentration: 7 per test group. Intermediate concentrations: 3 per test group.
- Biomass loading rate: Control and highest concentration: 0.20 g fish/litre, i.e. 7 fish per 8 litres of test medium. Intermediate concentrations: 0.23 g fish/litre, i.e. 3 fish per 3 litres of test medium.

TEST MEDIUM / WATER PARAMETERS

OTHER TEST CONDITIONS
- Adjustment of pH: no
- Photoperiod: 16 hours photoperiod daily with a light intensity in the range of 501 to 837 lux.
- Feeding: No feeding from 24 hours prior to the test and during the total test period.
- Introduction of fish: Within 20 minutes after preparation of the test media from a holding tank with comparable water quality parameters and pH and temperature differences between test and holding tank media of less than 1.0 unit and 1.0°C, respectively.

EFFECT PARAMETERS MEASURED
- Mortality and other effects: At 2.5, 24, 48, 72 and 96 hours following the start of exposure. In addition, every afternoon from day 0 to observe for any dead or severely distressed fish. Dead fish were removed when observed.

TEST CONCENTRATIONS
- Combined Limit/Range-finding study test concentrations: 1.0, 10 and 100 mg/L.

TERMINAL PROCEDURES
At the end of the test the surviving fish were rapidly killed by exposing them to ca. 1.2% ethylene glycol monophenylether in water.

Executive summary:

In a 96-h acute toxicity study conducted according to the OECD guideline 203 and GLP principles, the acute toxicity of 4-(cyclohexylamino)butane-1-sulfonic acid to carp (Cyprinus carpio) was investigated.


In an static combined limit/range-finding test, carp were exposed to an untreated control and to 1.0, 10 and 100 mg/L (control and 100 mg/L 7 fish per treatment and intermediate concentrations 3 fish per treatment). The measured concentrations of the highest tested concentration were at 92 and 96% relative to the nominal concentrations at the start and end of the test, respectively. Based on these results, effect parameters were expressed as analytically confirmed nominal concentrations. No mortality or other effects were observed in the control or at any of the concentrations tested.


In conclusion, the 96h-LC50 value for carp (Cyprinus carpio) exposed to 4-(cyclohexylamino)butane-1-sulfonic acid was beyond the range of concentrations tested, i.e. exceeded the concentration of 100 mg/L.
