The calculated model has a lower bound of -5.0 log units (all predictions lower than this value are set to -5.0). A dataset of compounds with experimental logP values has been built starting from the original dataset provided in EPI suite. The set has been processed and cleared from compounds that were replicated or that had problems with the provided molecule structure. The final dataset has 9,961 compounds.

For the two evaluated components of the substance, the Log Pow values as estimated by the VEGA/ALogP VEGA/Meylan-KOWWIN and SPARC models are reported. When relevant, the two most similar substances identified by VEGA are also included (using their CAS number), together with their similarity degree with the target component, experimental Log Pow values and the predictions obtained by the QSAR models.

The alcoholic moiety has two possible sites for esterification. In the table above, the structures originating from the mono esterification of the fatty acid in one of the two positions, are indicated with “-pos1” and “-pos2”.

Discussion

The applicability domain (AD) evaluation performed by the VEGA software suggests that the two evaluated components are not fully compliant with ALogP and KOWWIN applicability domains. The similarity degree indexes between the evaluated components and their two most similar substances identified be VEGA are high, whereas the indexes related to performances in predictions and to comparison between values predicted for the target molecules and the experimental ones available for their similar substances, are not optimal. Indeed, both VEGA models underestimate the Log Pow of the similar substances.

The applicability domain compliance with SPARC model cannot be clearly evaluated. However, SPARC models have been designed and parameterized to be applicable to any organic chemical structure. The partition coefficient model validation, performed on a dataset of 698 molecules, proved that the model can be used to provide reliable estimations. The predicted values for the two target components are therefore deemed to support the overall estimation of the Log Pow for the whole substance.

 

Conclusion

Considering the results obtained for the target components and the similar substances, a Log Pow value > 7 can be considered as a reasonable estimation for the whole substance.
